摘要
目的运用组织学和组织化学的方法观察苯甲酸雌二醇对绝经后骨质疏松模型大鼠子宫形态和子宫肥大细胞数量的影响。方法 3月龄健康雌性SD大鼠随机分为手术组(OVX组)和假手术组(Sham组),手术组实施双侧卵巢去除手术,手术后3个月将大鼠分为OVX组、OVX+E2组、Sham组,每组6只,OVX+E2组腹腔注射苯甲酸雌二醇20μg/(kg·d),每隔1天给药1次,连续给药28 d,OVX组和Sham组腹腔注射花生油作对照。给药结束后采集大鼠子宫常规石蜡包埋、切片,分别进行HE染色和改良甲苯胺蓝染色观察子宫形态并计数子宫横断面肥大细胞的数量。结果绝经后骨质疏松模型大鼠子宫明显萎缩,子宫腔径、子宫黏膜上皮厚度均较正常大鼠减小(P<0.01),给药苯甲酸雌二醇28 d后观察发现子宫腔径、子宫黏膜上皮厚度均恢复正常。对子宫肥大细胞的观察发现大鼠子宫肥大细胞均集中分布于子宫黏膜肌层、平滑肌束间结缔组织近小血管壁处,以微血管附近居多,子宫内膜内偶见。去卵巢后大鼠子宫肥大细胞较正常组显著增多(P<0.01),给药苯甲酸雌二醇后子宫肥大细胞数量趋于正常。结论提示卵巢雌激素在局部免疫调节中发挥重要作用。
Objective In this study, the effect of estradiol benzoate on the morphology of uterus and the number of mast cells in the uterus in rat postmenopausal osteoporosis model was observed using histology and histochemistry method. Methods Three- month-old SD rats were randomly divided into operation group ( OVX group ) and sham operation group ( Sham group ). After 3 months of the operation, the rats were divided into OVX group, OVX + E2 group, and Sham group, with 6 rats in each group. Estradiol benzoate (20 g/kg.d) was injected intraperitoneally in rats of OVX + E2 group, once every other day for 28 days. OVX group and Sham group were injected intraperitoneally with peanut oil as control. Rat uterus was collected after the end of administration. Tissues were then processed for paraffin embedding and sectioning by routine methods. HE and toluidine blue staining were used to observe the morphology of uterus and uterine mast cells. The number of mast cells in the uterus was counted. Results The uterine atrophied, and the diameter of the uterus and the epithelium thickness of the uterus decreased ( P 〈 0.01 ) in the postmenopausal osteoporosis model rats. The thickness of the uterine epithelium and the thickness of the uterine mucosa returned to normal after 28-day administration of estradiol benzoate. The uterine mast cells distributed in the smooth muscle layer of uterus mucosa, around the small blood vessels. They were less in the endometrium. The number of mast cells in the uterus of ovariectomized rats significantly increased in model rats ( P 〈 0.01 ), but it was back to normal after administration of estradiol benzoate. Conclusion Estrogen may play an important role in local immune regulation.
