摘要
目的:探讨补肾活血法对肾虚血瘀型大鼠雌孕激素及其受体的影响。方法:选动情前期大鼠按随机数字表法分为空白组、模型组、补肾活血方药低剂量组、高剂量组和五子组造肾虚血瘀模型,妊娠第4天处死大鼠,留取血清及子宫标本,放射免疫法测定血清雌二醇(E_2)、孕酮(P),免疫组化结合图像分析系统检测子宫雌激素受体(ER)、孕激素受体(PR)含量。结果:模型组E_2、P、ER、PR明显低于空白组;各治疗组E_2和P、高剂量组ER和PR、低剂量组和五子组的PR比模型组明显升高。多元相关性分析,ER、PR与E_2呈正相关,间质PR与P呈正相关。结论:补肾活血法可升高肾虚血瘀型大鼠E_2、P水平,调控ER、PR表达,利于子宫内膜成熟及胚泡着床。
Objective: To study the effects of tonifying the kidney and promoting blood circulation on estrogen and progesterone and their receptors in rats with kidney deficiency and blood stasis type. Method: the proestrus rats were randomly divided into blank group, model group, both low and high dose group of Bushen Huoxue recipe and the five sub group, made of kidney deficiency and blood stasis model, and killed on day 4 of pregnancy. Samples of their serum and uterine were collected for detecting serum estradiol (E2 ), progesterone (P) using radioimmunoassay; and endometrial expression contents of their receptors, ER and PR using immunohistochemistry and image analysis system. Results: E2 , P, ER and PR of the model group were significantly lower than blank group; E2 and P of the treatment group, ER and PR of high dose group, PR of low dose group and five sub group increased than the model group. Muhivariate correlation analysis, ER, PR and E2 were positively correlated, interstitial PR and P were positively. Conclusion: tonifying kidney and promoting blood circulation can improve E2, P level of kidney deficiency and blood stasis rats, and regulate the expression of ER, PR to improve endometrial maturation so as to conducive to blastocyst implantation.
