重组质粒pαACP-HSP70的特异性表达研究

摘要 目的观察重组质粒pαACP-HSP70在晶状体组织和非晶状体组织中的表达情况,探讨重组质粒pαACPHSP70的特异性表达。方法以晶状体组织作为A组,肺小细胞癌细胞A549、肝癌细胞7402等非晶状体组织作为B组,采用阳离子脂质体转染法将p CMV-HSP70质粒、pαACP-HSP70质粒导入A、B组后,通过荧光显微镜观察绿色荧光蛋白表达情况。结果转染p CMV-HSP70质粒到A、B组24 h后发现,两组均有绿色荧光;另外,转染pαACP-HSP70质粒24 h后仅在A组能观察到绿色荧光。结论重组质粒pαACP-HSP70不能在非晶状体组织表达,仅能在晶状体上皮细胞表达,具有组织特异性。 Objective To observe the expression of recombinant plasmid petACP-HSP70 in the lens and non-lens tissues for investigating its specific expression. Methods The lens tissue was taken as the group A and the non-lens tissue of small cell lung cancer A549 cells and liver cancer 7402 cells as the group B. pCMV-HSP70 and pctACP-HSP70 plasmids were transfected into the group A and B by adopting the cationic lipofectin transfection method. The expression of enhanced green fluorescent protein produced in the two groups was observed by fluorescence microscope. Results After 24 h of pCMV-HSP70 plasmid transfection into the group A and B ,the green fluorescence could be found in the two groups ,however,after transfecting p(xACP- HSP70 plasmid,the green fluorescence could be found only in the group A. Conclusion The pctACP-HSP70 recombination plasmid can not be expressed in non-lens tissue and only expressed in the lens epithelial cells with the tissue specificity.
