摘要
为研究AB-8大孔树脂对山丹鳞茎中总黄酮的纯化作用,采用静态和动态的吸附-洗脱试验,利用紫外可见分光光度计测量山丹鳞茎总黄酮的含量,研究不同的工艺条件对总黄酮纯化的影响。结果表明:原液浓度为3.165mg/m L,p H值为4,1.0m L/min的流速吸附,用70%的乙醇进行洗脱,可获得总黄酮纯度为35.13%。AB-8大孔树脂可较好的纯化山丹鳞茎中的总黄酮。
In order to study the purification effect of AB - 8 macroporous resin for total flavonoids from lilium pumi- lum bulb, the static and dynamic absorption - desorption methods are used, and the content of total flavonoids in samples is determined with UV spectrophotometer. The results show the optimum process for the adsorption that is the crude extract is adjusted to a concentration of 3. 165mg/mL, at pH 4, loading at a flow rate of 1.0 mL/min. The flowing of 70% ethanol aqueous solution through AB -8 column is found optimum for desorption. Under these conditions, the purity of total flavonoids is 35.13%. AB -8 macroporous adsorption resin is fit for purifying of total flavonoids from lilium pumilum bulb.
