摘要
目的:研究丹参多酚酸盐对阿霉素诱导的扩张型心肌病(DCM)大鼠心功能的作用及机制。方法:30只SD雄性大鼠分为对照组(10例)、阿霉素组(10例)和丹参多酚酸盐组(丹参多酚酸盐+阿霉素)(10例)。实验8周后行高频超声心动图检查,测定各项心功能指标后取心脏,行苏木精染色、masson染色及电镜观察。用TUNEL法检测各组大鼠心肌细胞凋亡,Western blot检测各组大鼠心肌细胞网腔钙结合蛋白(Calumenin)、内质网伴侣蛋白GRP78及凋亡因子chop、Bax、Bcl-2表达水平。结果:与阿霉素组比较,丹参多酚酸盐组大鼠心脏的收缩与舒张功能明显改善(P<0.01);心肌细胞凋亡明显减少(P<0.01);心肌细胞Calumenin表达增多,而GRP78及促凋亡因子chop、Bax表达减少(P<0.01),抑制凋亡因子Bcl-2表达增多(P<0.01)。结论:丹参多酚酸盐可改善DCM大鼠病理改变及心功能,减少DCM大鼠心肌细胞凋亡,上述作用可能通过增加心肌细胞Calumenin表达,缓解内质网应激,减少心肌细胞凋亡来实现。
Objective:To investigate the effect and mechanism of salvianolate on cardiac function in rats with dilated cardiomyopathy induced by adriamycin.Method:Sprague-Dawley male rats were randomly divided into control group(n=10),adriamycin group(n=10),salvianlate group(n=10).All the rats were sacrificed after measuring the cardiac function indexes by high frequency echocardiography on 8th week.The morphological changes of myocardial cells was observed by using HE staining,masson staining and electron microscopic.The apoptosis was detected by TUNEL method and the expression of calumenin,endoplasmic reticulum chaperone GRP78,chop,Bax,Bcl-2 was monitored by Western blotting.Result:1 Compared with adriamycin group,the cardiac systolic and diastolic function was improved significantly in salvianlate group.2 Compared with adriamycin group,the amount of myocardial apoptosis in salvianlate group decreases obviously.3 Compared with adriamycin group,the expression of calumenin was improved and endoplasmic reticulum chaperone GRP78,chop,Bax,Bcl-2 was decreased in salvianlate group.Conclusion:Salvianlate can improve the pathological changes and cardiac function of dilated cardiomyopathy rats and decrease the myocardial cell apoptosis in rats with dilated cardiomyopathy.These effects may be achieved by increasing the expression of Calumenin protein in myocardium,relieving the endoplasmic reticulum stress and reducing the apoptosis.
