摘要
目的检测IL-10、IL-17及IL-18在肝细胞肝癌中的表达特点,为肝细胞肝癌的诊断与治疗提供实验依据。方法采用RT-PCR方法检测肝癌以及癌旁组织中IL-10、IL-17及IL-18mRNA水平的表达,采用Western Blot、免疫组化方法检测肝癌以及癌旁组织中IL-10、IL-17及IL-18蛋白水平的表达;采用ELISA检测肝癌细胞株BEL-7402、Hep G2中IL-10、IL-17及IL-18的释放水平。结果 IL-10、IL-17在肝癌组织及细胞株中高表达,与血清AFP检测结果呈正相关;IL-18在肝癌组织及细胞株中不表达或者少量表达,与血清AFP检测结果呈负相关。结论 IL-10、IL-17的过表达和IL-18的沉默与肝细胞肝癌的发生密切相关,IL-10、IL-17的过表达可作为肝细胞肝癌诊断的重要标志。
Objective To investigate the expression characteristics of IL-10,IL-17 and IL-18 in hepatocellular carcinoma( HCC) to provide experimental base for the diagnosis and treatment of HCC. Methods The mRNA expressions of IL-10,IL-17 and IL-18 in hepatocellular carcinoma tissues were detected by RT-PCR. The protein expressions of IL-10,IL-17 and IL-18 were detected by Western Blot and immunohistochemical method. The releasing of IL-10,IL-17 and IL-18 in BEL-7402,Hep G2 cells were detected by ELISA. Results The IL-10 and IL-17 were over expressed in HCCs tissues and cells,and they had positive relationship with serum AFP. The expression of IL-18 in HCCs tissues and cells was down-regulated and had negative relationship with AFP. Conclusion The over expression of IL-10 and IL-17 and down regulation of IL-18 were closely related with the pathogenesis of HCC. The IL-10 and IL-17 could be used as important markers for the diagnosis of HCCs.
