摘要 目的以恶性胶质瘤细胞系(U87)为细胞模型研究实时无标记细胞分析技术(real-time cellular analysis,RTCA)在细胞系培养质量控制中的应用。方法记录三株不同来源U87细胞系72小时实时细胞增殖曲线,并结合显微镜下细胞的形态学特征,判断细胞系是否发生变异。结果与购自协和的细胞株U87-1相比,U87-2、U87-3显微镜下形态学发生了明显的改变,同时细胞增殖曲线结果也显示出明显的变异。结论实验室存在细胞系在培养过程中发生变化的现象,若使用发生变化的细胞,将影响后续的实验结果。通过RTCA技术在细胞系培养时进行质量监控非常必要。 Objective To investigate the application of real-time cellular analysis( RTCA) in quality control of cell culture by using malignant glioma cell lines( U87) as a model. Methods The real-time cell proliferation curve of three malignant glioma cell lines of U87 were recorded for 72 hours and the cell lines mutation were analyzed according to the curve and the morphological characteristics. Results Compared with U87-1 purchased from Xiehe,both U87-2 and U87-3 cell lines had obviously change in microscopic morphology and cell proliferation curve. Conclusion Cell lines could change during the cultivating. Using these changed cell lines will affect the results of the following experiment. It is necessary to use real-time cellular analysis for the quality control of cell culture.
