BAF/GM细胞株法在生长激素生物测定中的应用

摘要 目的探讨BAF/GM细胞株法测定正常儿童及特发性身材矮小患儿生长激素生物活性中的应用,为特发性身材矮小的病因学研究提供一定的实验依据。方法采用BAF/GM细胞株法建立体外生物测定系统,测定正常组儿童及特发性身材矮小患儿生长激素生物活性,同时采用免疫放射分析法测定其免疫活性。通过加入胰岛素、胰岛素样生长因子等评估该检测方法的稳定性,分析其对生长激素的阻断作用。结果 (1)生长激素刺激BAF/GM细胞株的增殖呈剂量依赖性。(2)两组儿童血清生长激素生物活性和免疫活性比较差异均无统计学意义(P1=0.393,P2=0.54)。(3)游离胰岛素样生长因子在浓度>30ng/mL时刺激细胞增殖,且增殖呈剂量依赖性,其余生长因子对细胞增殖无影响。结论 BAF-GM细胞株法测定生长激素生物活性具有较好的敏感性与特异性,是分析血清样本中生长激素的生物活性水平的一种切实可行方法。 Objiective To establish a new bioassay method for GH bioactivity.By using this methodology,to evaluate the availability of the method through an intensive GH bioactivity assay of samples in idiopathic short stature(ISS).Methods Establish BaF/GM cell line,then evaluate the availability of the method through an intensive GH bioactivity assay on ISS and control,as well as measure its immuno-activity by radiometric assay(IRMA).And evaluate the stability of the testing method by test insulin and insulin-like growth factor 1(IGF-1)to observe its inhibition on growth hormone.Results(1)The hGH stimulated BaF/GM cell proliferation in a dose-dependent manner and this bioassay was sensitive even at-0.020ng/mL.(2)There were no statistically significant differences between the two groups in bioactivity and immunoactivity(P1=0.393,P2=0.54).(3)IGF-1increased cell proliferation at a concentration of more than 30ng/mL and stimulated the cell proliferation in a dose-dependent manner.Conclusion Thus the newly developed system is a specific,sensitive,easy and fast bioassay system for GH determination and it is useful for evaluation of GH bioactivity in the variety of endocrine states.
