桥本甲状腺炎患者外周血IL-14的表达及意义 被引量:1

Expression and significance of IL- 14 in PB of patients with Hashimoto's thyroiditis

摘要 目的检测桥本甲状腺炎患者外周血中白细胞介素-14(IL-14)的表达水平,研究IL-14与甲状腺过氧化物酶抗体(TPOAb)及甲状腺球蛋白抗体(Tg Ab)的相关性。方法选择55例桥本甲状腺炎患者作为实验组,26例健康体检者作为对照组。用TRIzol试剂提取实验组患者及对照组体检者的外周血中单个核细胞总RNA,逆转录为c DNA,采用实时荧光定量PCR(real-time PCR)检测IL-14 mRNA表达水平;采用酶联免疫吸附法(ELISA)检测外周血中IL-14蛋白质表达水平。结果实验组患者外周血中IL-14表达高于对照组(P<0.05);此外,IL-14表达水平与TPOAb、Tg Ab分别呈正相关(P<0.05)。结论桥本甲状腺炎患者外周血中IL-14表达增高,且与TPOAb、Tg Ab水平呈正相关,IL-14可能参与桥本甲状腺炎的发病。 Objective To detect the expression level of serum interleukin- 14( IL- 14) in peripheral blood( PB) of patients with Hashimoto's thyroiditis,and to analyze its relationship with thyroid peroxidase antibody( TPOAb) and thyroglobulin antibody( Tg Ab). Methods 55 patients with Hashimoto's thyroiditis were enrolled as experiment group. 26 normal cases were enrolled as control group. Total RNA in PBMCs from the experiment group and the control group were extracted by using TRIzol method. Then,the extracted total RNA was reversely transcribed into c DNA. Subsequently,real- time PCR was used to assay IL- 14 mRNA expression level. Enzyme- linked immunosorbent assay( ELSIA) method was used to detect IL- 14 protein expression level in PB. Results Serum IL- 14 expression levels in the experiment group was higher than that in the control group( P 〈0. 05). Serum IL- 14 level was positively related to serum TPOAb and Tg Ab( P 〈0. 05). Conclusion Expression level of serum IL- 14 increased in patients with Hashimoto's thyroiditis. Furthermore,IL- 14 expression level was positively related to serum TPOAb and Tg Ab levels,respectively. It is suggested IL- 14 might be involved in the pathogenesis of Hashimoto's thyroiditis.
