高效液相色谱-电喷雾质谱分析白杨素在大鼠体内的代谢产物 被引量:4

摘要 目的探讨白杨素在大鼠体内的代谢产物。方法给药组以白杨素2 mg/m L的溶液灌胃雄性SD大鼠,同时空白组大鼠以等体积的溶剂灌胃。灌胃24 h后分别取两组大鼠的尿液和粪便,同时分别于给药后0.5、1、2、4、6 h对两组大鼠进行眼眶取血。应用液质联用推测化合物的可能结构。结果在大鼠尿、粪便、血浆检测到原型成分白杨素M1(白杨素),在大鼠粪、胆汁中检测到M2(白杨素硫酸结合物);此外,在大鼠尿液、血、胆汁等样品中检测到代谢成分M3-M4(白杨素葡萄糖醛酸结合物)。结论白杨素在大鼠体内的主要代谢方式为Ⅱ相代谢,产物是葡萄糖醛酸结合物与硫酸结合物。 Objective To study the metabolism of chrysin in rats. Methods Drug group was given intragastric administration with chrysin solution 2 mg/m L, at the same time blank group was given intragastric administration with equal volume of solvent. Urine and feces of the two groups were taken after all rats were administered 24 hours later, at the same time at half an hour, 1 hour, 2 hours, 4 hours, 6 hours of the medicine given, all rats were performed orbital blood. Through LC-MS, according to the information, the possible structures of the compounds were speculated. Results In rat urine, feces and plasma, prototype components chrysin M1 was detected, M2 was detected in fecal bile(chrysin sulfate conjugates); in addition, M3-M4 metabolites were detected in rat urine, blood and bile samples(chrysin glucuronide). Conclusion The main metabolism method of chrysin in rats is phase Ⅱ metabolism, the metabolites are glucuronic acid conjugate and sulfuric acid conjugate.
