摘要
目的建立RP-HPLC法同时测定神康宁丸中斯皮诺素、迷迭香酸、紫草酸和丹酚酸B 4种成分的含量。方法采用高效液相色谱法(HPLC),以Platisil ODS(250mm×4.6mm,5mm)为分析柱,乙腈-体积分数为0.1%的磷酸溶液为流动相进行梯度洗脱,流速:1.0mL·min-1,柱温:40℃,检测波长:340nm,进样量:20μL。结果斯皮诺素、迷迭香酸、紫草酸和丹酚酸B的质量浓度分别在1.032~10.32、0.460 0~4.600、1.736~17.36和21.12~211.2mg·L-1内与峰面积呈良好的线性关系(r≥0.999 6,n=6)。斯皮诺素、迷迭香酸、紫草酸和丹酚酸B平均回收率分别为100.9%、101.3%、100.4%和99.8%(RSD≤1.5%,n=9)。结论所建立的HPLC法可用于神康宁丸的质量控制。
Objective To develop an RP-HPLC wavelength switching method for simultaneous determination of spinosin,rosmarinic acid,lithospermic acid and salvianolic acid B in Shenkangning pills.Methods The HPLC separation was achieved on a Platisil ODS(4.6 mm×250 mm,5μm)column at 40 ℃.The mobile phase consisted of acetonirile-0.1% phosphoric acid solution with gradient elution at a flow rate of1.0mL·min-1.Results The linear range was 1.032-10.32 mg·L-1(r=0.999 9,n =6),0.46-4.6 mg·L-1(r=0.999 8,n=6),1.736-17.36 mg·L-1(r=0.999 6,n=6),21.12-211.2 mg·L-1(r=1.000,n=6),respectively.The recovery was 100.9%(RSD=0.7%,n=9),101.3%(RSD=1.5%,n=9),100.4%(RSD=1.0%,n=9),99.8%(RSD=1.2%,n=9)respectively.Conclusions The method is simple,accurate and reproducible and can be used for quality control of Shenkangning pills.
