产木聚糖酶菌株的筛选、鉴定及酶学性质研究 被引量:14

Screening and Identification of Xylanase-producing Microorganisms and Study on Enzyme Properties

摘要 【目的】从不同来源的样品中筛选产木聚糖酶的菌株,并对其进行菌种鉴定和酶学性质研究。【方法】以自制木聚糖为唯一碳源,采用平板透明圈方法筛选产木聚糖酶的菌株,结合菌株的形态特征和16S r DNA序列分析鉴定菌种,并对菌株所产木聚糖酶进行酶学性质研究。【结果】筛选到的44株有透明圈的菌株中27株具有产木聚糖酶能力,其中B659菌株产酶能力最高;经鉴定B659菌株为同温层芽孢杆菌;B659菌株所产木聚糖酶的最适反应温度55℃,最适反应p H值6.5,在37℃、p H 8~10的条件下保温2 h,仍具有78%以上的酶活力以及较好的耐碱性。10 mmol/L K+和1mmol/L Fe^(2+)对该酶具有促进作用,Mn^(2+)、Cu^(2+)和Co^(2+)对该酶具有明显的抑制作用。【结论】筛选到具有较好耐碱性的木聚糖酶高产菌株——同温层芽孢杆菌B659。 [Objective] Xylanase-producing strains were screened from different sources of samples. The strain which had largest xylanase yield was identified and its xylanase properties were studied. [Methods] With self-made xylan as the only carbon source, the xylnase-producing strains had been screened according to the transparent zone on the plate. The strain was identified based on the morphological characteristics observation and 16 S rDNA sequence analysis. [Results]The 27 of 44 strains which showed transparent zones had xylanse-producing ability and B659 was the highest among them. B659 was identified as Bacillus stratosphericus. The optimal temperature and pH of B659 xylanse was 55 ℃ and6.5. 78% of xylanse activity was retained after 2 h incubation at 37 ℃ and pH 8-10 and it performed great alkali resistance. Besides, 10mmol/L K+and 1 mmol/L Fe^2+displayed promotion effect to xylnase ability while Mn^2+, Cu^2+and Co^2+manifested obvious inhibition. [Conclusion] Bacillus stratosphericus B659 with high ability to produce alkali-resistance xylanase was screened.
