摘要 目的建立浓百精简合剂的质量标准控制方法。方法采用薄层色谱法对浓百精简合剂中黄芩、五味子2味中药进行定性鉴别;采用RP-HPLC法测定浓百精简合剂中厚朴酚及和厚朴酚的含量,色谱柱为Agilent Eclipse XDB-C18(4.6 mm×150 mm,5μm),流动相为甲醇-0.1%磷酸水(65∶35,v∶v),流速为1.0 m L/min,检测波长294 nm。结果薄层色谱法能检测出浓百精简合剂中的黄芩、五味子,特征明显,专属性强;RPHPLC可测定厚朴酚、和厚朴酚的含量。厚朴酚在0.050 8~0.355 6μg范围内线性关系良好(r=0.999 3),和厚朴酚在0.025 15~0.176 05μg范围内线性关系良好(r=0.999 3)。厚朴酚加样回收率为98.28%,RSD=1.25%;和厚朴酚为98.62%,RSD=0.76%。结论通过制定该质量标准,确保对浓百精简合剂进行快速地定性及定量分析,实验重复性良好,结果稳定、可靠,能够控制浓百精简合剂的质量。 Objective To improve the quality criteria of simple Nongbai mixture. Methods TLC was used to make qualitative determination of Chinese magnoliavine fruit and scutellaria baicalensis in simple Nongbai mixture and RP-HPLC was used to determine the content of its main constituents of magnolol and honokiol. For the determination of magnolol and honokiol,the chromagraphic column was Agilent Eclipse XDB-C18( 4. 6 mm × 150 mm,5 μm); the mobile phase consisted of methanol-0. 1% phosphoric acid water( 65∶ 35 v ∶ v); the flowrate was 1 m L / min; the peaks were monitored at 294 nm. Results Chinese magnoliavine fruit and scutellaria baicalensis in simple Nongbai mixture could be identified by TLC. The contents of the calibration curve of magnolol showed good linerity( r = 0. 999 3) within the range of 0. 050 8 ~ 0. 355 6 μg; the calibration curve of honokiol showed good linearity( r = 0. 999 3) within the range of 0. 025 15 ~ 0. 176 05 μg. The average recovery of magnolol was 98. 28% and the RSD was 1. 25%. The average recovery of honokiol was 98. 62% and the RSD was 0. 76%. Conclusion The present method is stable,accurate and highly repetitive,which can be used for the quality control of this preparation.
