HPLC测定不同产地、不同品种绞股蓝中芦丁和槲皮素的含量 被引量:12

Content Determination of Rutin and Quercetin in Gynostemma pentaphyllum from Different Producing Areas by HPLC

摘要 目的:建立高效液相色谱同时测定不同产地、不同品种绞股蓝药材中芦丁和槲皮素含量的方法。方法:ZORBAX SB-C18色谱柱(4.6 mm×150 mm,5μm),流动相乙腈(A)-0.2%磷酸溶液(B)梯度洗脱,流速1.0 m L·min-1,柱温30℃,检测波长360 nm,进样量10μL。结果:芦丁在2.535~10.14μg线性良好(R^2=0.999 2),平均回收率为98.72%,RSD 1.2%;槲皮素在0.018 1~0.144 8μg线性良好(R^2=0.999 3),平均回收率98.40%,RSD 1.7%。结论:该方法简便可行、重复性好,适用于绞股蓝药材2种有效成分的同时测定,可为绞股蓝药材的质量控制提供参考依据。 Objective: To establish a determination method for different varieties of rutinand quercetin in Gynostemma pentaphyllum from different producing areas. Method: HPLC was performed on ZORBAX SB-C18column( 4. 6 mm × 150 mm,5 μm),eluting in a gradient mode with the mobile phase of acetonitrile( A)-0. 2%H3PO4as( B) at a flow rate of 1. 0 m L·min- 1. The column temperature was 30 ℃. The detection wavelength was360 nm. The injection volume was 10 μL. Result: The linearity of rutin was good within the range of 2. 535-10. 14 μg( R^2= 0. 999 2),and the average recovery was 98. 72%( RSD 1. 2%). The linearity of quercetin was good within the range of 0. 018 1-0. 144 8 μg( R^2= 0. 999 3),and the average recovery of 98. 40%( RSD 1. 7%).Conclusion: The established method is simple,feasible and highly reproducible,and can be used for determining rutin and quercetin in G. pentaphyllum and providing reference for quality control of G. pentaphyllum.
