摘要
采用真空-超声提取联用高效液相色谱法测定了槐米中的槲皮素,并与常压超声提取和氮气条件下超声提取的结果进行了比对。测定结果表明:真空-超声提取法的提取率远高于常压超声提取和氮气条件下超声提取的结果,且真空条件是决定槐米中槲皮素提取率的关键因素,而室温提取时氧气的存在并不产生明显的影响。扫描电镜结果表明:真空条件下提取后的样品表面有更加明显的溶蚀和破坏,这种变化有利于溶解并释放槐米中的槲皮素,从而提高提取率。
Quercetin in sophora flower bud was determined by vacuum ultrasonic extraction coupled with high performance liquid chromatography method.The extraction efficiency was compared with ultrasonic extraction under the conditions of atmospheric pressure and nitrogen,respectively.The results show that the extraction efficiency by vacuum ultrasonic extraction method is much better than that by ultrasonic extraction under the conditions of atmospheric pressure and nitrogen,respectively.Vacuum condition is the key factor to extract quercetin,and the oxygen existed at room temperature does not influence the extraction efficiency.Scanning electron microscope results show that sample surface is significantly by eroded and destroyed after vacuum ultrasonic extraction,which is conducive to dissolve and release quercetin in sophora flower bud and to improve the extraction efficiency.
