脾肾阳虚型溃疡性结肠炎大鼠模型的建立及四神丸加味的反证效果 被引量:9

摘要 目的:探索脾肾阳虚型溃疡性结肠炎大鼠模型的建立方法并观察四神丸加味对模型的反证效果。方法:将40只雄性SD大鼠随机分为正常对照组、三硝基苯磺酸(TNBS)模型组、病证模型组、以方测证组。正常对照组给予普通喂养;TNBS模型组采用TNBS与乙醇复合物灌肠进行造模;病证模型组给予番泻叶、腺嘌呤分阶段灌胃、外因干预、TNBS与乙醇复合物灌肠进行造模;以方测证组在复合因素建立脾肾阳虚型溃疡性结肠炎模型后予四神丸加味煎剂灌胃。实验结束后观察各组大鼠症状、体征及结肠病理改变,并检测血清中IL-6、IL-8和TNF-α水平。结果:病证模型组大鼠出现嗜睡懒动,反应迟钝,蜷卧扎堆,毛发疏松粗糙、晦暗无光泽,眯眼弓背,肛周污秽,便形稀溏甚则黏液血便等症状;大鼠体质量明显减轻,饮水量明显减少,肛温下降,显微镜下观察结肠黏膜呈缺损,可见水肿、充血、大量炎症细胞浸润,伴有溃疡灶形成,结肠组织损伤程度评分较高;病证模型组大鼠血清IL-6、IL-8、TNF-α水平均高于正常对照组,差异有统计学意义(P<0.05或P<0.01)。以方测证组大鼠体质量明显增加,饮水量明显增加,肛温较前升高,结肠黏膜见轻度充血、水肿,有少量炎性细胞浸润;以方测证组大鼠结肠组织损伤程度评分、血清中IL-6、IL-8、TNF-α的浓度均低于病证模型组,差异均有统计学意义(P<0.05或P<0.01)。结论:番泻叶、腺嘌呤分阶段灌胃结合环境因素干预、TNBS与乙醇复合物灌肠造模建立脾肾阳虚型溃疡性结肠炎大鼠模型是可行的。四神丸加味能改善溃疡性结肠炎脾肾阳虚型大鼠模型相关症状及体征。 Objective: To explore the establishment method of ulcerative colitis of spleen kidney Yang deficiency rats and the counterevidence with Supplemented Sishen Pill(四神丸). Methods: The 40 male SD rats were randomly divided into control group, trinitrobenzene sulfonic acid(TNBS) model group, disease and syndrome model group, prescriptions measured model group.The control group was given normal feeding; TNBS model group was induced through enema by compound of TNBS and alcohol; The disease and syndrome model group was firstly given Fanxieye(folium sennae) and adenine through gavage in stages simultaneously with external intervention, and then induced as TNBS model group did. The prescriptions measured model group was treated by Supplemented Sishen Pill decoction through gavage on the basis of disease and syndrome model group. The symptoms, signs and colon pathological changes of rats were observed after the end of the experiment, and the levels of IL-8, TNF-αand IL-6 in serum were detected. Results: The disease and syndrome model rats appeared sleepiness lazy, unresponsive, lay curled up and get together, loosed hair and rough, squinting camponotus, crissum filth, form loose even mucous bloody stool and other symptoms; the body weight and the water consumption of the rats were significantly decreased, and the anus temperature decreased;under the microscope, there was a defect in the colon mucosa, which was found to have edema, congestion,inflammatory cell infiltration, and the formation of ulcer lesions; serum levels of IL-6, IL-8 and TNF-αwere significantly higher than those in normal control group, and the difference was statistically significant(P〈0.05 or P〈0.01). The body weight and water consumption of the rats in the test group was significantly increased after treatment, the anus temperature increased, and the colon mucosa was mild congestion and edema, and a small amount of inflammatory cell infiltration; The degree of colon injury score and serum levels of IL-6, IL-8 and TNF-αwere decreased, and the difference was statistically significant(P〈0.05 or P〈0.01). Conclusion: Fanxieye(folium sennae), adenine stages gastric perfusion, environmental factors intervention combined with TNBS and Ethanol compound enema modeling with Yang deficiency of spleen and kidney ulcer colitis rat model established is feasible. Supplemented Sishen pill can improve the symptoms related with ulcerative colitis of spleen kidney yang deficiency animal model and signs.
