丹参注射液对氧诱导幼鼠视网膜增生变性的保护作用 被引量:2

摘要 目的探讨丹参注射液对氧诱导幼鼠视网膜凋亡相关基因的表达及其外周血血管内皮生长因子(VEGF)的影响,以明确其抑制视网膜增生变性的作用机制。方法将100只出生7 d的SD幼鼠随机分为对照组、模型组及丹参注射液4,2,1 g/kg组,每组20只。对照组幼鼠生活在正常氧环境下,其余4组幼鼠均制成视网膜病变模型。模型组给予腹腔注射生理盐水,丹参注射液4,2,1 g/kg组分别给予腹腔注射相应剂量的丹参注射液,连续注射5 d后处死幼鼠,经右侧眼球取静脉血检测VEGF浓度,取出眼球用RT-PCR法检测视网膜中bax与bcl-2 mRNA表达水平。结果模型组幼鼠视网膜组织中bax mRNA表达水平明显低于对照组(P<0.05),而血清VEGF浓度与视网膜组织中bcl-2 mRNA表达水平均明显高于对照组(P均<0.05)。丹参注射液2 g/kg组血清VEGF浓度及视网膜组织中bcl-2 mRNA表达水平均明显低于丹参注射液1 g/kg组(P均<0.05),高于丹参注射液4 g/kg组(P均<0.05);视网膜组织中bax mRNA表达水平明显高于丹参注射液1 g/kg组(P<0.05),低于丹参注射液4 g/kg组(P<0.05)。bax mRNA表达水平与bcl-2 mRNA表达水平间存在显著负相关关系(r=-0.653,P<0.05)。结论丹参注射液能抑制氧诱导的幼鼠视网膜细胞增生。 Objective It is to explore the effect of Danshen injection on the expression of apoptosis correlated gene and vascular endothelial growth factor( VEGF) in the serum in the neonatal rats with oxygen induced retinopathy,and to research the mechanism of inhibiting hyperplasia. Methods 100 neonatal rats( 7- day old) were randomly divided into five groups:control group,model group,Danshen injection groups( 4,2,1 g / kg),every group included 20 rats. The rats in control group lived in the environment with normal oxygen. The rats in other groups were used to establish models of retinopathy of prematurity. Model group was given normal saline,Danshen injection groups were given 4,2,1 g / kg Danshen injection respectively.After intervention for 5 days,the rats were killed to draw their blood to detect the concentration of VEGF,and to get their retina to determine the expression levels of bax and bcl- 2 through RT- PCR. Results The level of bax mRNA in retina tissue in model group was lower while that of bcl- mRNA was higher than that in control group( P〈0. 05). The content of serum VEGF and the expression of bcl- 2 mRNA in 2 g / kg Danshen injection group were lower than that in 1 g / kg Danshen group( P〈0. 05),but higher than that in 4 g/kg Danshen injection group( P〈0. 05). The expression of bax mRNA in retina tissue in 2 g / kg Danshen injection group were higher than that in 1 g / kg Danshen group( P〈0. 05),but lower than that in 4 g / kg Danshen injection group( P〈0. 05). Bax mRNA expression was negatively related with bcl- 2 mRNA( r =- 0. 653,P〈0. 05). Conclusion Ginkgo biloba extract can inhibit the hyperplasia induced by oxygen.
