Box-Behnken响应面法优化裂殖壶菌油脂胞内转乙酯化工艺条件的研究

摘要 为了优化裂殖壶菌油脂胞内转乙酯化工艺条件,以DHA酯化率为指标,在单因素实验基础上采用Box-Behnken响应面实验设计,考察了无水乙醇用量、浓硫酸用量、反应温度和反应时间对裂殖壶菌油脂胞内转乙酯化的影响。结果表明:针对0.5 g干菌粉,得到最优的胞内转乙酯化工艺条件为无水乙醇用量11.76 m L、浓硫酸用量0.41 m L、反应温度75.5℃、反应时间2.24 h,在此条件下DHA酯化率达到85.19%。 The effects of dosages of anhydrous ethanol and concentrated sulfuric acid,reaction temperature and reaction time on the intracellular ethyl esterification transfer of oil from Schizochytrium sp. were investigated by Box-Behnken response surface design with esterification rate of DHA as index based on the single factor experiment so as to optimize the intracellular ethyl esterification transfer conditions of oil from Schizochytrium sp.. The results showed that the optimal intracellular ethyl esterification transfer conditions of 0. 5 g dry bacterial powder were obtained as follows: dosages of anhydrous ethanol and concentrated sulfuric acid 11. 76 m L and 0. 41 m L respectively,reaction temperature 75. 5 ℃ and reaction time 2. 24 h. Under the optimal conditions,the esterification rate of DHA reached 85. 19%.
