摘要
[目的]观察大株红景天注射液抗心肌缺氧/复氧损伤的保护作用,并探讨其抗心肌缺氧/复氧可能的机制。[方法]原代培养的心肌细胞随机分为5组:空白对照组;缺氧/复氧组;缺氧/复氧分别加大株红景天注射液5、10、20 m L/L组。加药孵育12 h后,缺氧培养9 h,随后常氧培养2 h。噻唑蓝(MTT)法检测细胞增殖,CMH2DCF-DA荧光探针检测细胞内活性氧簇(ROS)的生成量,JC-1荧光探针检测细胞内线粒体膜电位,以及测定细胞内超氧化物歧化酶SOD活力及其基因表达量。[结果]大株红景天注射液增加了缺氧/复氧损伤心肌细胞的活力,减少细胞内ROS的生成,抑制细胞内线粒体膜电位的降低,同时增加了心肌细胞内SOD的活力及其基因表达量。[结论]大株红景天注射液对缺氧/复氧(H/R)损伤的心肌细胞有保护作用。
[Objective] To investigate the effect of sofren injection on primary cardiomyocytes damaged by hypoxia-reoxygenation and to research signal pathways involved in the mechanism of sofren injection. [Methods] The cell viability was measured by MTT. Primarily cultured cardiomyocytes were pretreated with different concentrations of sofren injection, cultured in hypoxia for 9 h and reoxygenation for2 h. The cell viability was measured by MTT. The ROS generation and mitochondrial membrane potential in the cell were measured by CMH2DCF-DA and JC-1 fluorescent probe, the vitality and gene expression of superoxide dismutase(SOD). [Results] Compared with hypoxiareoxygenation group, pretreated with different concentrations of sofren injection significantly attenuated model-induced cell viability and mitochondrial membrane potential loss, dramatically decreased the ROS generation. The SOD vitality and gene expression were increased by sofren injection. [Conclusion] Sofren injection can protect rat cardiomyocytes from the injury induced by hypoxia-reoxygenation.
