摘要
目的了解葛根牛磺酸复合片对小鼠酒精性肝损伤的作用。方法将60只健康ICR小鼠,随机分为3个剂量组(250、500、1 500mg/kg)和溶剂组、模型组,3个剂量组按不同剂量给予葛根牛磺酸片,溶剂组和模型组给予纯净水,灌胃量均为20mL/kg b.wt.,连续30d后3个剂量组和模型组均一次灌胃给予50%乙醇14mL/kg b.wt.,禁食16h,进行组织病理学检查及丙二醛(MDA)、谷胱甘肽(GSH)、甘油三酯(TG)含量测定。结果与溶剂组相比,模型组小鼠含脂滴的肝细胞数量增加,肝匀浆中MDA含量升高,肝匀浆中TG含量增高,GSH活性降低(P<0.01),表明建模成功。与模型组比较,葛根牛磺酸复合片低、高剂量组小鼠肝细胞脂肪变性有不同程度减轻(P<0.05);高剂量组小鼠肝匀浆中MDA值降低(P<0.05);中、高剂量组小鼠肝匀浆中GSH含量均有增加(P<0.05);中、高剂量组小鼠肝匀浆中TG含量均有下降(P<0.01)。结论葛根牛磺酸复合片对酒精性肝损伤有一定的辅助保护功能。
Objective To investigate preventive effect of Puerariae Radix and Taurin(CPRT)compound on alcoholic liver injury.Methods A total of 60 ICR mice were randomly divided to 3dose groups(250,500,1 500mg/kg b.wt.),solvent group and model group.Mice in dose groups were treated with different doses of CPRT while mice in solvent group and model group were treated with distilled water by intragastric administration of 20 mL/kg b.wt..After 30 days of treatment,mice in dose groups and model group were given 50%ethanol(14mL/kg b.wt.)followed by 16 hfasting.Surgical-pathological examination was taken,MDA,GSH and TG concentrations were determined.Results Compared with solvent group,mice in model group had increased number of liver cells with lipid droplets,MDA and TG in liver homogenate increased,while GSH activity decreased(all P〈0.01).All above results demonstrated liver injury model was successfully established.For mice treated with low and high CPRT doses,fatty degeneration of liver cells were alleviated at certain degrees(all P〈0.05).MDA concentration of high dose group decreased significantly(P〈0.05);GSH concentration increased significantly(P〈0.05)of mice in medium and high dose groups while TG concentration decreased significantly(P〈0.01).Conclusion CPRT could protect alcoholic liver injury at certain degrees.
