摘要
采用聚酰胺-大孔树脂联用法纯化大叶藻黄酮,对其纯化工艺进行优化。样液以聚酰胺吸附,水洗除杂后置于大孔树脂柱上部,以70%(v/v)的乙醇洗脱。通过单因素和正交试验优化纯化的最佳工艺,确定最佳纯化工艺条件是样液与大孔树脂的比为4:1(v/m),样液与聚酰胺的比为16:1(v/m),洗脱体积为6 BV,洗脱流速为2 BV/h,在此条件下黄酮纯度由(14.97±0.51)%增加到(71.14±0.79)%。该方法纯化工艺简单,具有良好的应用前景。
The process parameters of the purification of Zostera marina L.flavonoids by polyamidemacroporous resin were studied.Polyamide which adsorbed with sample was washed with water first to eliminate the impurities.Then,put polyamide in macroporous resin column above.The flavonoids were enriched by eluting with 70% ethanol.The optimized process for the purification of Zostera marina L.flavonoids was explored on the basis of single factor experiments and orthogonal experiments.The optimized technology conditions to purify Zostera marina L.flavonoids were as follow:the ratio of sample and macroporous resin was 4:1,the ratio of sample and polyamidewas 16:1,eluting the column with 6 BV at a flow rate of 2 BV/h.After the purification,the purity of flavonoids was increased from(14.97±0.51)% to(71.14±0.79)%.The method is more convenient,which has a good application foreground.
