摘要
以红米为原料,采用8种大孔树脂对红米色素进行静态吸附和解吸试验,筛选出合适的树脂型号,确定用AB-8对红米色素进行纯化,得到AB-8吸附红米色素平衡时间为4h,解吸终点时间为120min。通过单因素和正交试验得到了最佳动态吸附工艺参数,即上柱液流速1.0ml/min、上柱液温度20℃、上柱液pH3.0、上柱液质量浓度1.5mg/ml;最佳动态解吸条件为:乙醇体积分数75%,pH1.5,洗脱流速为1.0ml/min,解吸率最高。在此条件下,纯化的色素呈暗红色,经过纯化后色价为9.79,比未纯化红米色素提高了30%。
With red rice as raw material,using 8kinds of macroporous resin for red rice pigment static adsorption and desorption test to select the suitable resin,AB-8resin was determined for purification of red rice pigment.The equilibrium time of AB-8adsorption red rice pigment was 4h,the desorption end time was 120 min.Through single factor and orthogonal experiment,the optimal dynamic adsorption process parameters were as:the upper column liquid flow rate of 1.0ml/min,the column temperature of 20℃,pH3.0and the column concentration of 1.5mg/ml;the optimal dynamic desorption conditions were ethanol concentration 75%,ethanol pH 1.5,elution flow rate of 1.0ml/min,desorption rate was the highest.Under the selected condition,the pigment was dark red,and the color value was 9.79 after purification,increased by 30%.
