摘要
利用SMART RACE技术克隆获得泥蚶卵黄原蛋白(Vitellogenin Vg)部分c DNA序列,长为1 411 bp,包含1 362 bp的开放阅读框,编码454个氨基酸.空间表达结果发现泥蚶Vg m RNA主要在卵巢和消化腺中表达.在1,3,6,9μg·L-1铅(Pb)胁迫28 d后检测发现,前3个质量浓度组泥蚶消化腺中的Vg表达量呈先抑制后升高趋势;而在9μg·L-1 Pb暴毒组中,Vg在整个实验过程中均被诱导表达,表达量呈先升高后降低趋势.4个实验组中,泥蚶消化腺Vg表达量均在21 d达到最大值(P<0.01).故泥蚶消化腺Vg表达量和Pb胁迫具有一定的时间效应和剂量效应,推测其抗氧化剂功能在Pb胁迫引起的氧化损伤中起作用.
Using the SMART RACE method, Vitellogenin(Vg) part c DNA sequence is obtained. The fragment c DNA of Vg gene is 1 411 bp, which consists of an open reading frame of 1 362 bp encoding a polypeptide of 454 amino acids. Homology alignment results reveal that the similarity of deduced amino acid sequence with other bivalve shellfish is higher than 49%. The results of tissue-specific expression by real time PCR show that Vg of Tegillarca granosa in breeding period is mainly expressed in ovarian and digestive gland. After challenged by 1, 3, 6, 9 μg·L-1 concentration of Pb for 28 days, the results of relative expression in digestive gland of Tegillarca granosa in non-breeding period of Vg m RNA is obtained showing that the first three groups are first slightly reduced followed by a significant increase. Throughout the experiment, Vg m RNA is used as the induced expression and the expression quantity is obtained after the first rise followed by decrease under stress of the concentrations of 9 μg·L-1 Pb. Vg expression reaches highest at 21 d under Pb stress(P0.01). In addition, Pb induces Vg expression with the relationship between dose-effect and time-effect. It is speculated that the antioxidant functions play a role in the oxidative damage caused by Pb stress.
