摘要
研究制备了禽白血病病毒抗原检测能力比对样品。禽白血病病毒(ALV)经56℃1h灭活后,采用商品化试剂盒进行初步定值,并用DMEM稀释制备出强、弱阳性和阴性样品,分装后保存。采用ELISA方法进行比对样品均匀性和稳定性检验,并发送32个实验室进行测试,所有参试实验室的检测结果符合定值范围。结果显示制备的比对样品均匀性和稳定性良好,满足检测能力验证要求。
Proficiency testing samples for avian leukosis virus（ALV）were prepared in this study.Cell culture of ALV was inactivated at 56 ℃ for 1 h and was identified as primary samples.p27 antigen of primary samples was determined by IDEXX ALV Ag test kit.Primary samples were diluted by DMEM to prepare strong positive samples,weak positive samples and negative samples and homogeneity and stability of the samples were verified by ELISA kit.5 samples with strong positive,weak positive and negative samples were treated as one proficiency testing sample.32 Proficiency Testing samples were sent to 32 laboratories in China.All the testing results of the samples meet the requirement of given value.This study showed that the prepared samples could be used in Proficiency Testing for ALV antigen test.
