摘要
目的建立茯苓中去氢土莫酸和茯苓酸含量的测定方法。方法采用紫外波长转换检测的RP-HPLC法。色谱柱为ZORBAX Eclipse XDB-C18柱(4.6mm×150 mm,5μm),流动相为乙腈-0.05%磷酸水溶液(75∶25),检测波长为0~5 min,241 nm(去氢土莫酸),5~16 min,210 nm(茯苓酸),流速为1.0 ml·min-1,柱温为30℃。结果去氢土莫酸和茯苓酸分别在25.5~255.0μg·ml-1(r=0.9999)和11.16~111.6μg·ml-1(r=0.9999),浓度与峰面积呈良好的线性关系;方法回收率分别为100.4%(RSD=2.8%)、99.1%(RSD=2.9%)。结论 HPLC法快速、准确、选择性好、灵敏度高,适合茯苓中去氢土莫酸和茯苓酸的含量测定。
Objective To establish an HPLC method for the determination of dehydrotumulosic acid and pachymic acid in Poria.Methods A RP-HPLC method with variable UV wavelengths had been developed,including 241 nm( 0 ~ 5 min) for dehydrotumulosic acid and 210 nm( 5 ~ 16 min) for pachymic acid. Chromatographic separation was carried out with a ZORBAX Eclipse XDB-C18column( 4. 6 × 150 mm,5 μm) and a mixture of acetonitrile and 0. 05% phosphate acid( 75∶ 25) as mobile phase.The flow rate was 1. 0 ml · min-1,and the column temperature was 30 ℃. Results The linear range was 25. 5 μg·ml-1~255. 0 μg·ml-1( r = 0. 9999) for dehydrotumulosic acid,and 11. 16 μg·ml-1~ 111. 6 μg·ml-1( r = 0. 9999) for polyporenic acid. The average recoveries of these compounds were 100. 4%( RSD = 2. 8%) and 99. 1%( RSD = 2. 9%),respectively.Conclusion The proposed HPLC method is rapid,accurate,selective,sensitive,and suitable for the determination of the content of pachymic acid and dehydrotumulosic acid in Poria.
