MT1-MMP在病毒性心肌炎小鼠中的表达及三七总皂甙的干预作用 被引量:4

摘要 目的观察膜型基质金属蛋白酶-1(MT1-MMP)在病毒性心肌炎小鼠中的表达及意义。方法选用近交系4~6周龄雄性BALB/C小鼠,采用随机数字表法分为3组:空白对照组、心肌炎组、治疗组。于病毒接种后第8、15、21天分别采血后处死小鼠并留取心脏标本。分别采用RT—PCR法及免疫组化法检测心肌MT1-MMP的mRNA及蛋白表达,光镜观察心肌组织病理改变。结果心肌炎组和治疗组各时间点心肌细胞MT1-MMP的mRNA含量明显高于空白对照组(P〈0.01),治疗组在第15、21天明显低于心肌炎组(P〈0.05或0.01);心肌炎组和治疗组各时间点心肌细胞MT1-MMP免疫组化染色呈强阳性表达,空白对照组呈弱阳性。与心肌炎组比较,治疗组心肌细胞MT1-MMP表达在第15、21天降低,差异均有统计学意义(P〈0.05或001)。结论MT1-MMP可能参与了病毒性心肌炎小鼠的发病,中药三七总皂甙可能通过降低MT1-MMP的表达起治疗作用。 Objective To investigate the expression of membrane type 1 matrix metalloproteinase (MT1-MMP) in myocardium of mice with viral myocarditis, and the effect of panax notoginsenosides (PNS) on MT1-MMP. Methods Sixty eight inbred male Balb/C mice of 4-6 weeks were divided randomly into 3 groups: the mice in blank group (group C) were inoculated intraperitoneally (i.p.) with free virus 1640 culture solution 0.1ml on day 0, the mice in VMC group (group V) were inoculated i.p. with 0.1ml Tissue Culture Infectious Dose 50 (TCID50) of 10-436/ml Coxsackie viruses B3 (CVB3) on day 0, the mice in treatment group (group T) were inoculated i.p with Coxsackie viruses B3 and PNS 180mg/kg. The animals were sacrificed on d8, d15, and d21 after infections. The hearts tissue slides were HE stained for histological changes; and immunohistochemically (IHC) stained for MT1-MMP protein expression. The MT1-MMP mRNA expression was detected by RT-PCR method. Results The expressions of MT1-MMP mRNA in groups V and T were higher than group C in all time points (P〈0.01); the expression was lower in group T than that in group V at d15 and d21 (P〈0.0 5 or 0.01). The IHC staining showed that myocardial cells had strong expression of MT1-MMP in groups V and T and mild expression in the group C. Compared with group V, the IHC OD values of group T was significantly lower at d15 and d21 (P〈0.0 5 or 0.01). Conclusion MT1-MMP may be involved in the pathogenesis of viral myocarditis in mice. Traditional Chinese medicine PNS may have some therapeutic effect by reducing the expression of MT1-MMP.
