摘要
利用扩增片段长度多态性(Amplified fragment length polymorphism,AFLP)分子标记技术,对26份鸡冠花种质资源进行亲缘关系探讨。结果表明:1)采用8对引物组合共扩增出8 063条谱带,其中多态性条带为7 959条,多态性条带比率为98.71%,平均每对引物扩增出1 007.9个位点和994.9个多态性位点。26份种质的相似性系数居于0.264 2~0.664 8,平均值为0.464 5,每个位点的有效等位基因数为1.078 0,平均多样性指数为0.208 7,遗传相似性系数最高的是1和3号,达到0.664 8,最低的是13和24号,为0.264 2。根据遗传相似性系数进行聚类分析,在遗传相似性系数0.46处切割时,可将图中26个鸡冠花种质资源分为3个类群:24号杂交种粉色头状鸡冠花单独聚为一类,8号多头红色鸡冠花单独聚为一类;其他24个鸡冠花材料聚为一类。根据不同的花型和花色,24个鸡冠花材料又可分成6个小类群。分子聚类结果和形态聚类结果较为一致。24号杂交粉色头状鸡冠花和8号多头鸡冠花‘威信’,与其他24个鸡冠花材料的亲缘关系较远;来源于相同地区的种质资源,如19和25号同为来自于甘肃的品种亲缘关系较近;来自科研所自育品种16和23号单独聚为一类,与其他种质的亲缘关系较远。
The research on genetic diversity and relationship of 26 samples of Celosia cristata,Celosiaplumosa,Celosia argenteaL.Provides knowledge support for germplasm innovation and variety right protection.Among 8 063 bands detected by 8elective primer pairs,7 959 bands(98.71%)were polymorphic.On average,each primer combination amplified 1 007.9loci and 994.9polymorphic loci.Genetic similarity of 26 cockscomb samples ranged from0.264 2to 0.664 8with an average of 0.464 5.The number of effective alleles for each locus was 1.078 0,and the average index of genetic diversity was 0.208 7.The highest genetic similarity(0.664 8)was found between sample#1and #3,and the lowest(0.264 2)was found between # 13 and # 24.Cockcomb germplasm was divided into3 groups in term of a threshold of genetic similarity of 0.64.sample #24,a hybrid variety,was placed in a single member group Ⅰ.Cockcomb germplasm coded 8,which is a multi-receptacle variety,was classified into group Ⅱ.Other 24 samples were classified into groupⅢ,which could be further divided into 6subgroups with different shape and color of receptacles.Cluster analysis shows that samples#24and#8were separated genetically from other samples.Furthermore,varieties from same seed company(Gansu Seed Company),such as #19and #25,had close genetic relationship.Varieties bred by Beijing Institute of Landscape and Architecture,such as #16and #23,were also clustered in one group.It indicates that these varieties have distinct genetic from other varieties.
