摘要
为了研究蛋白质磷酸化修饰状况在体细胞被诱导为多能性干细胞过程中发挥的作用,试验采用TiO_2富集磷酸化肽段技术和高通量质谱技术,对第三代牛胎儿成纤维细胞的磷酸化修饰蛋白质组进行定性检测和生物信息学分析。结果表明:在牛胎儿成纤维细胞样品中共鉴定出363种磷酸化蛋白,共502个磷酸化位点。这些磷酸化蛋白参与多种细胞信号传导通路,在代谢进程和细胞定位等生物学进程中发挥重要作用。说明对牛胎儿成纤维细胞磷酸化修饰蛋白质组的定性研究可为进一步探讨牛体细胞重编程为诱导性多能干细胞的机理和提高重编程效率奠定理论基础。
To study the role of the modification status of protein phosphosrylation status in the process of somatic cells induced to pluripotent stem cells,a technique for Ti O2 enrichment of phosphorylated peptides and high- throughput mass spectrometry technique were used for qualitative detection and bioinformatics analysis of the phosphoproteome from the third- passage bovine fetal fibroblasts( BFFs). The results showed that a total of 363 phosphoproteins with 502 phosphorylation sites were identified in the samples of BFFs. These phosphoproteins participated in multiple- cell signal transduction pathways,and played an important role in the metabolic processes,cellular localization,and other biological processes. The results indicate that the qualitative research on the phosphoproteome of BFFs may lay a theoretical foundation for further exploring the mechanism of somatic cell reprogramming to the induced pluripotent stem cells and improving the efficiency of reprogramming.
