摘要
目的:研究血管紧张素1型受体(AT1R)基因A1166C多态性与贵州地区布依族、汉族原发性高血压(EH)的相关性。方法:在贵州省独山县人民医院病房及门诊收集的EH确诊病例为高血压组,布依族77例,汉族74例;门诊体检选取健康成年人群为正常对照组,布依族64例,汉族78例。采用PCR扩增技术及限制性酶切法(PCR-RFLP),检测布依族、汉族高血压组及正常对照组的外周血白细胞DNA中AT1R基因A1166C的多态性,统计分析布依族、汉族高血压组和正常对照组中A1166C基因型及等位基因频率的分布情况。结果:布依族高血压组的AA、AC和CC基因型频率分别为93.5%、6.5%和0,而在正常对照组中分别为95.3%、4.7%和0,两组进行对比差异无统计学意义(P=0.728);布依族高血压组中A与C等位基因频率分别为96.8%、3.2%,而在正常对照组中分别为97.7%、2.3%,两组比较差异也无统计学意义(P=0.732)。汉族高血压组的AA、AC和CC基因型频率分别为87.8%、12.2%和0,而在正常对照组中分别为97.4%、2.6%和0,两组进行对比差异有统计学意义(P=0.022);汉族高血压组中A与C等位基因频率分别为93.9%、6.1%,而在正常对照组中分别为98.7%、1.3%,两组比较差异有统计学意义(P=0.025),高血压组的AC基因型与C等位基因频率显著增高。结论:在贵州地区,AT1R基因A1166C多态性与布依族人群的EH无显著相关性,而与汉族人群的EH具有显著相关性,AC基因型和C等位基因可能是高血压发生的危险因素。
Objective:To investigate the correlation between AT1 R A1166Cgene polymorphism and essential hypertension in Buyi and Han from Guizhou province.Method:We collected cases with essential hypertension as the high blood pressure group,comprising 77 cases of Buyi,and 74 cases of Han ethnic group.And healthy adults were employed as the normal control group,including 64 cases of Buyi,and 78 cases of Han nationality.We used PCR amplification technology and restriction enzyme digestion method(PCR-RFLP)to detect A1166 Cgene polymorphisms of peripheral blood leukocyte DNA in Buyi and Han patients with essential hypertension(Hypertension group)and people with normal blood pressure(Normal control gronp)from Guizhou province.Result:Buyi hypertension group AA,AC and CC genotype frequencies were 93.5%,6.5% and 0,while in the normal control group was 95.3%,4.7% and 0,with no difference(P=0.7280.05).Buyi hypertension group A and C allele frequencies were 96.8%,3.2%,and in the normal control group was 97.7%,2.3%,with no difference(P=0.7320.05).Han hypertension group AA,AC and CC genotype frequencies were 87.8%,12% and 0,while in the normal control group was 97.4%,2.6%and 0,with significant difference(P=0.0220.05).Han hypertension group A and C allele frequencies were 93.9%,6.1%,and in the normal control group was 98.7% and 1.3%,with significant difference(P=0.0250.05).AC genotype and C allele frequency was significantly higher in hypertensive group.Conclusion:In Guizhou province,there was no significant correlation between AT1 R A1166C gene polymorphism and essential hypertension in Buyi people.However,it has significant correlation between AT1R A1166 Cgene polymorphism and essential hypertension in Han people.AC genotype and C allele may be a risk factor for hypertension.
