摘要
以细菌污染的晋南1号单倍体组培枣苗为材料,以乙醇、氯化汞和超声波为主要试验因素,采用随机区组试验设计,探讨超声波辅助氯化汞消毒拯救污染组培苗及外植体消毒的优化方法。结果表明,拯救污染组培苗的优化组合消毒方法是70%乙醇浸泡30 s+0.1%氯化汞浸泡4 min+超声(28 Hz,25℃)15 min。该方法较之常规乙醇-氯化汞化学消毒方法,接种的外植体污染率降低了68.41%,其成活率提高了75.72%,增殖程度提高了4.41倍。该超声波辅助氯化汞消毒的理化新方法,降低了外植体消毒中氯化汞的使用浓度和消毒时间,减少了消毒对环境的污染,能极大地提高消毒效果,可扩展推广到所有植物组织培养中外植体的消毒。
To obtain Zizyphus jujube mill,Jinnan 1,tissue culture seedling contaminated with bacteria,methods were studied by using the ethanol,mercuric chloride and ultrasonic as primarily test factors together with the random block design of experiment. The results showed that optimal combined disinfection method for saving pollution tissue culture seedling was as follows samples soaked in 70% ethanol for 30 s,then in 0. 1% mercuric chloride for 4 min and treated with ultrasonic( 28 Hz,25℃) for 15 min. Compared with the conventional ethanol-mercury chloride chemical disinfection method,pollution rate of inoculation explants with this method was reduced by 68. 41%,the survival rate increased by75. 72%,the proliferation degree increased by 4. 41 times. This new method of physical and chemical disinfection showed good feasibility.
