摘要
研究响应面法与酶法联用工艺提取拐枣中的活性物质拐枣多糖及其抗氧化活性。采用辅助热水提取法提取拐枣多糖,以Box-Behnken试验设计结合响应面分析法优化了提取工艺条件,确定最佳工艺参数。结果显示,在酶解温度为50℃,酶解浓度为0.06 g和p H为5.0的条件下,拐枣多糖的最高含量为19.79%。拐枣多糖具有明显的抗氧化作用和清除自由基的作用,当质量浓度为5.0 mg·m L-1时,FRAP值为1.91,对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基、1,1-二苯基-2-苦基肼基自由基、羟基自由基和超氧阴离子自由基的清除率分别达到69.63%、74.10%、44.13%和73.79%。
Polysaccharides were extracted from Hovenia acerba fruits with the help of hot water after enzymolysis and response surface methodology. The antioxidant activity of the extracted polysaccharides was determined. The main extraction conditions were optimized using response surface methodology based on Box- Behnken design. An extraction condition with the enzymolysis temperature of 50℃ and enzymolysis concentration of 0.06 g at p H of 5.0 was optimal. The extraction yield under the optimized conditions was 19.79%. The polysaccharides extracted from Hovenia acerba fruits showed antioxidant capacity and free radical scavenging capacity. At the concentration of 5.0 mg·m L-1, the FRAP value was 1.91. the ABTS+·, DPPH racial, superoxide anion radical and hydroxyl free radical scavenging rates were 69.63%, 74.10%, 44.13% and 73.79%, respectively.
