摘要
采用ISSR标记技术研究11个肉桂(Cinnamomum cassia)家系之间的亲缘关系,研究结果表明:用12条ISSR引物对11份肉桂供试材料检测出条带154条,其中多态性条带80条,比例为51.95%;11份肉桂家系供试材料的等位基因平均数Na为1.5195,有效等位基因Ne为1.3604,基因多样性指数即平均期望杂合度He为0.2046,Shannon信息指数I为0.2999。聚类分析显示,11份供试材料间的遗传距离在0.1307~0.3548之间,说明其遗传亲缘关系相对较近,在遗传距离为0.2495处可较明显将11份供试材料分为6个类群;在遗传距离0.2920处分为2个类群,种间各自聚类显著,可区分清化桂和西江桂。
ISSR was used to study genetic relationship of 11 Cinnamomum cassia genealogies. Results showed that 12 ISSR primers which were used in 11 tested materials of Cinnamomum cassia generated 154 legible bands, of which 80 were polymorphic, occupying for 51.95%. The mean number of alleles (Na) was 1. 519 5. The mean effective number of alleles (Ne) was 1. 360 4. The Nei's gene diversity (He) was 0. 204 6. The Shannon' s information index (I) was 0. 299 9. The genetic distance varied from 0. 130 7 to 0. 354 8 among 11 tested materials by cluster analysis, which indicated that genetic relation- ship was close. 11 Cinnamomum cassia genealogies could he classified into 6 groups when genetic distance was 0. 249 5. Two groups were classified when genetic distance was 0. 292 0, with significantly interspecifie clustering, which could separate Cinnamomum cassia var. macrophylla and Cinnamomum cassia from Xijiang.
