摘要
目的观察Yes相关蛋白(YAP)在乳腺癌及癌旁组织中的表达情况,探讨其与乳腺癌侵袭转移的关系。方法收集我院2008年1月至2009年1月手术治疗的乳腺癌患者资料以及石蜡标本共177例。采用免疫组织化学的方法观察YAP、基质金属蛋白酶(MMP)-2在乳腺癌组织及癌旁组织的表达情况,探讨YAP与乳腺癌侵袭转移的关系。结果(1)YAP在乳腺癌组织中的表达率仅为29.8%(28/94),显著低于YAP在癌旁组织中的表达率(74.7%,62/83),差异有统计学意义;发现YAP的低表达与组织学类型(x^2=20.897,P〈0.01)和肿块的大小(x^2=6.623,P〈0.05)有关;YAP在乳腺癌组织内的表达主在胞质,而在正常乳腺上皮组织中的表达主在细胞核内。(2)乳腺癌组织中YAP与MMP-2的表达呈负相关(r=-0.253,P〈0.05)。YAP的低表达与组织学类型肿块大小有关,MMP-2的高表达与淋巴结是否发生转移有关。结论YAP在乳腺癌组织可能扮演抑癌基因的角色,其可能抑制肿瘤的侵袭转移。
Objective To observe the expression of Yes - associated protein (YAP) in breast cancer tissues and adjacent tissues. To explore the relationship between YAP and breast cancer invasion and metastasis. Methods Collected the surgical treatment of breast cancer patients' clinical data and par- affin specimens from January 2008 to January 2009 in our hospital. The results showed that we collected a total of 177 cases, with 94 cases of breast cancer tissue and 83 cases adjacent to carcinoma. Through im- munohistochemical methods and explore the relationship between YAP and Breast Cancer invasion and me- tastasis. Results ( 1 ) The expression rates of YAP in breast cancer tissues is 29. 8% (28/94) which is significantly lower than that of in adjacent tissues to carcinoma (74. 7%, 62/83 ) and the difference has statistically significant. In breast cancer tissues, YAP protein is mainly located in the cytoplasm, while in normal breast epithelium, YAP protein is mainly located nucleus. The lack or low of YAP in breast cancer tissues is correlation with histological type (x^2 = 20. 897, P 〈 0. 01 ) and tumor size (x^2 = 6. 623, P 〈 0. 05 ). ( 2 ) YAP expression has negatively correlated with the expression of matrix metalloproteinase - 2 ( MMP - 2, r = - 0. 253, P 〈 0. 05 ). Conclusion YAP functions as a tumor suppressor gene in breast cancer, which may suppress tumor invasion and metastasis.
