摘要
为了促进废弃油脂发酵制备生物表面活性剂的研究及生产,对生物表面活性剂产生菌进行了筛选,并研究优良菌种的发酵特性。采用平板分离法和摇瓶发酵筛选法,从长期受餐厨废油脂污染的土壤中获得性能优良的菌种WO-S14,经菌体形态特征、生理生化特征和16S r DNA等方面分析,该菌种被鉴定为嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia)。以豆油为唯一碳源,菌种WO-S14的发酵产物经薄层层析显色分析和傅里叶红外光谱分析,被确定为脂肽类生物表面活性剂。通过对菌种WO-S14的发酵条件进行初步研究,最佳的初始p H、温度和摇瓶转速分别被确定为7.0、30℃和200 r/min,生物表面活性剂的最大产量达0.92 g/L。
Biosurfactant is a kind of high molecular compounds produced by microbial metabolism, which mainly consists of glycolipids,peptide lipids,lipoproteins,fatty acids,phospholipids,neutral lipids and high molecular polymers. The present study was to examine the screening and isolation of the biosurfactant-producing strain utilizing waste grease as raw material. The fermentation characteristics of the target strain were also studied. Through plate separation and shaking flask tests,an excellent biosurfactant-producing strain of WO-S14 was isolated from soil which was contaminated by waste cooking oil in the long time. According to the morphological features, physiological and biochemical characteristics and 16 S r DNA sequence analysis, the strain of WO-S14 was identified as Stenotrophomonas maltophilia while soybean oil as the sole carbon source. The results of thin-layer chromatography(TLC) and Infrared spectrometric identification(IR) showed that the fermentation product of WO-S14 was determined as lipopeptide bio-surfactant. The optimal fermentation conditions of WO-S14 were as follows: initial p H value of 7.0,temperature of 30 ℃ and rotation speed of 200r/min. Under the optimal conditions,the yield of bio-surfactant could reach up to 0.92 g/L.
