摘要 目的观察不同浓度葡萄糖对体外培养的人成骨细胞的影响,探讨糖尿病致骨质疏松症可能的发病机制。方法将4种不同浓度葡萄糖(5.5,10,20,40 mmol/L)的培养基培养人成骨细胞,培养3~12 d。观察葡萄糖对细胞增殖、凋亡及对细胞分泌碱性磷酸酶(ALP)和骨钙素(BGP)能力的影响。结果在一定浓度范围内,葡萄糖浓度越高,细胞增殖越明显,细胞分泌ALP和BGP的能力下降,细胞凋亡率增加。结论高糖环境对人成骨细胞分泌能力有抑制作用。 Objective: In this research,we took an insight into the influence of different glucose concentrations on human Osteoblast cell,hoping to give an explanation to the mechanism of the osteoporosis( OP) caused by diabetes mellitus( DM). Methods: Four culture media of different glucose concentration( 5. 5,10,20 or 40 mmol/L) were set up for this study. Human Osteoblast cell was cultured in these media successively for 3 ~ 12 d,and then the proliferation rate of cells,secretion of ALP and BGP,percentage of apoptosis cells were observed and measured. Results: within a certain glucose concentration,the elevated glucose concentration prompted the proliferation of Osteoblast cells,but the activities of Osteoblast cells were not correspondent with the cell number. On the contrary,the secretion of ALP and BGP of the Osteoblast cells were decreased. The apoptosis of the cells was promoted. Conclusion: From our study,we confirmed the hyperglycemia can inhibit the secretion ability of Osteoblast cells.
