摘要
以 2 ,3 二氢 2 ,2 二甲基 7 苯并呋喃酚为反应原料经两步化学反应合成了 2种克百威半抗原 :6 [[(2 ,3 二氢 2 ,2 二甲基 7 苯并呋喃基氧 )羰基 ]氨基 ]己酸 (BFNH)和 4 [[(2 ,3 二氢 2 ,2 二甲基 7 苯并呋喃基氧 )羰基 ]氨基 ]丁酸 (BFNB)。通过活性酯法和混合酸酐法将半抗原与载体蛋白 (BSA、OVA)偶联制备了克百威的人工抗原。利用BFNH BSA和BFNB BSA分别免疫日本大耳白家兔获得了 2种抗克百威的多克隆抗体 :抗BFNH BSA抗体和抗BFNB BSA抗体 ,并用间接竞争ELISA方法测定了 2种抗体的效价。
To produce high titre polyclonal antibodies(PAbs) to the pesticide carbofuran, two compounds named 4-[[(2,3-dihydro-2,2-dimethyl-7-benzofuranyloxy)carbonyl]-amino]butanoic acidbutanoic acid and 6-[[(2,3-dihydro-2,2-dimetyl-7-benzofuranyl-oxy)carbonyl]-amino]hexanoic acidhexanoic acid were synthesized, conjugated to bovine serum albumin (BSA) with the modified active ester method, and artificial antigen of Carbofuran was prepared and then they were as immunizing haptens in rabbits. 2 kinds of anti-carbofuran antibodies were obtained. The method of the indirect competitive ELISA for carbofuran was established and used to determine titre of the antibodies. The antibodies had high titre and specificity for Carbofuran.
