趋化性细胞因子RANTES在大鼠肺纤维化模型的表达及其意义

摘要 目的研究RANTES(regulated upon activation normal T cell expressed and secreted facter)在肺纤维化发病过程中的表达特征及其与病程的相关性。方法 64只健康成年雌性大鼠随机分为模型组和对照组,每组32只。模型组气管内一次性注射博来霉素构建特发性肺纤维化模型,对照组仅注射生理盐水。于第1、3、7、14天分别处死大鼠,显微镜下观察肺组织病理改变;碱水解法检测大鼠肺组织匀浆中的羟脯氨酸含量;ELISA测定肺组织中的RANTES蛋白水平;Realtime PCR检测肺组织中的RANTES m RNA水平。结果病理分析及羟脯氨酸检测显示肺纤维化模型构建成功;第7、14天模型组羟脯氨酸含量明显升高,且显著高于对照组(P<0.01);模型组的大鼠肺组织RANTES蛋白及m RNA表达水平在第3天迅速增高并达到高峰,此后逐渐下降,但维持至第14天时仍显著高于对照组(P<0.01)。结论 RANTES在早期肺纤维化模型大鼠肺组织中显著升高,其变化早于肺组织羟脯氨酸含量的变化及病理改变,可能是肺组织纤维化的早期标记。 Objective To investigate the expression of regulated upon activation normal T cell expressed and secreted factor(RANTES) during pathogenetic process of pulmonary fibrosis and its relevance to the disease course. Methods Sixty four healthy adult female rats were randomly divided into Model Group and Control Group. Model Group received single intratracheal instillation of bleomycin while Control Group received single intratracheal instillation of normal saline only. The rates in both groups were sacrificed on Day 1, 3, 7, and 14, respectively. Pathological changes in lung tissues were observed under microscope; alkalinehydrolysis was used to detect the content of hydroxyproline in lung tissue homogenate in rats; in the, the two group were sacrificed on day separately. Elisa was used to measure the expression of RANTES; and Real-time PCR was used to detect the expression of RANTES m RNA in rat lungs. Results Pathological analysis and hydroxyproline detection showed that lung fibrosis mode was successfully built; On Day 7 and 14, the content of hydroxyproline in Model Group was promptly increased and reached the peak, and then gradually decreased, but still significantly higher than that in Control Group on Day 14(P〈0.01). Conclusion The expression of RANTES in early pulmonary fibrosis model was significantly increased. The change was earlier than the content change of hydroxyproline and pathological changes in lung tissues, and may be early marker of pulmonary fibrosis.
