金属钴、铬离子对成骨细胞增殖及Ⅰ型胶原蛋白表达功能的影响 被引量:6

摘要 目的观察钴、铬离子对小鼠成骨细胞(MC3T3-E1)的细胞毒性及Ⅰ型胶原蛋白合成的影响,探讨钴、铬离子对骨代谢功能的影响机制。方法钴、铬离子分别与小鼠成骨样MC3T3-E1细胞体外培养,噻唑蓝(MTT)法测定细胞活力。ELISA法检测培养上清液中Ⅰ型胶原蛋白含量、RT-PCR检测成骨细胞Ⅰ型胶原蛋白m RNA的表达。结果 MTT结果显示,与对照组相比,钴、铬离子能明显抑制成骨细胞的细胞活力;成骨细胞暴露在钴铬离子下,与对照组相比,24h、48h后:Ⅰ型胶原蛋白的分泌分别下降45.3%、49.2%(P<0.05);Ⅰ型胶原蛋白m RNA表达分别下降26.8%、32.6%(P<0.05)。结论钴、铬离子可抑制成骨细胞Ⅰ型胶原蛋白的分泌及其m RNA的表达,提示其对MC3T3-El细胞可能存在细胞毒性。 Objective To investigate the cytotoxicity of cobalt(Co^2+) and chromium(Cr^3+) ions on mice osteoblast(MC3T3-E1)and the effects on the secretion of collagen-Ⅰ from osteoblast under the stimulation of Co^2+ and Cr^3+ ions. Methods The osteoblast was cultured in vitro. The cell vitality was assured by MTT test. The ELISA and RT-PCR methods were applied to detect the collagen-Ⅰ on gene and protein levels at 24 and 48 hours after co-culture respectively. Results Compared to the control,MTT test demonstrated that Co^2+ and Cr^3+ ions can obviously decrease the cell viability of osteoblast. When the osteoblasts were exposed to Co^2+ and Cr^3+ ions for 24 and 48 hours,the releasion of collagen-Ⅰ decreased by 45.3% and 49.2% as compared with the control(P〈0.05);the expression of collagen-Ⅰ m RNA decreased by 26.8% at 24h,and 32.6% at 48h as compared with the group A(P〈0.05). Conclusion Co^2+ and Cr^3+ ions can inhibit the expression and release of collagen-Ⅰ from osteoblast,and may have a cytotoxic effect on osteoblasts.
