意大利牛舌草中芦丁的分离及定量分析研究 被引量:4

摘要 目的确定意大利牛舌草中芦丁的分离方法并建立用于测定意大利牛舌草中芦丁含量的HPLC方法。方法采用硅胶柱色谱对意大利牛舌草乙酸乙酯萃取物进行分离纯化。建立测定意大利牛舌草中芦丁含量的反相高效液相色谱法,色谱条件:色谱柱Kromasil C18(5μm,250mm×4.6mm);以乙腈-0.2%磷酸水溶液进行线性梯度洗脱:0~9min 83%~80%B,9~20min 80%~68%B,20~40min 68%~40%B,40~45min 40%~83%;流速1.0mL/min;检测波长354nm,柱温30℃。结果通过分离鉴定在意大利牛舌草乙酸乙酯萃取物中得到单体化合物1,1 H-NMR与13 C-NMR结果提示其化合物1为芦丁。在1.64×10^(-3)~5.255×10^(-2 )mL/min范围内芦丁浓度与其峰面积呈良好的线性关系(r=0.999 7),且不同批次的意大利牛舌草中的芦丁含量最低,为(1.299 7±0.014 1)mg/g。结论首次从意大利牛舌草中分离得到芦丁,所建立的用于测定芦丁含量的HPLC法简便、准确,重现性好,结果准确可靠,为提高意大利牛舌草的质量标准奠定了试验基础。 Objective To separate and identify the contents of rutin extracted fromAnchusa italica Retz..Methods The ethyl acetate extracts were isolated and separated by silica gel,the structure was identified by ~1 H-NMR and ~(13) C-NMR.The contents identification of rutin was performed by RP-HPLC.The conditions of HPLC was as follows:Kromasil C_(18)(5μm,250mm×4.6mm)columns was used and gradient elution solvents was the mixture of acetonitrile and 0.2% phosphoric acid,the flow rate was 1.0mL/min,the detective wave length was at 354 nm,column temperature was 30℃.Results Rutin was obtained and identified from Ethyl acetate extracts.There was a linear relationship between concentration of rutin and peak area in the range of 0.00164mL/min~0.05255mL/min,and the content of rutin in different batches Anchusa italica Retz.is 1.2997±0.0141 mg/g.Conclusion Rutin were firstly isolated from Anchusa italica Retz.The identification method using HPLC was simple,accurate and reliable,which lays the experimental fundatioin for the establishment of quality standard of Anchusa italica Retz.
