摘要
目的:观察真武汤对心力衰竭大鼠血清ST2及IL-33的影响,并探讨其作用机制。方法:以80只健康雄性Wistar大鼠为研究对象,除空白组10只外,其余70只通过冠脉结扎手术,术后结合冷水力竭式游泳,复制心力衰竭动物模型,把造模后存活的50只大鼠随机分为真武汤高剂量组、中剂量组、低剂量组、西药组和模型组5组,分别予以真武汤、卡托普利、生理盐水灌胃,疗程15 d。用酶联免疫吸附试验法(ELISA)测定大鼠血浆ST2、白细胞介素-33(IL-33)水平。结果:中药高剂量组、西药组FS值与EF值与模型组比较显著升高(P<0.01),中药高剂量组、西药组大鼠FS值、EF值与正常组比较差异无统计学意义(P>0.05);模型组ST2、IL-33水平均显著升高(P<0.01),中药高剂量组、中剂量组与西药组ST2、IL-33水平显著降低(P<0.01),西药组与中药高剂量组、中剂量组比较差异无统计学意义(P>0.05),中药高剂量组、中剂量组IL-33水平较正常组无统计学意义(P>0.05)。结论:真武汤能够明显降低心力衰竭大鼠血清ST2、IL-33水平,说明真武汤通过调控IL-33/ST2L信号通路改善心室重构,这可能是真武汤治疗心力衰竭的机制之一。
Objective:To observe the effect of Zhenwu Decoction on serum ST2 and IL-33 in heart failure rats,and to explore its mechanism of action. Methods:In 80 healthy male Wistar rats as the research object,except the blank group(10 rats),the remaining 70 only by ligation of coronary artery,postoperative combined with cold water swimming exhaustion,replication of animal model of heart failure,modeled after the survival of 50 rats were randomly divided into two groups:Zhenwu Decoction high dose group,middle dose group,low dose group,western medicine group and model group 5 group,and were treated with Zhenwu Decoction and captopril,saline gavage treatment course was 15 days. ST2 and IL-33 levels were measured by enzyme-linked immunosorbent assay(ELISA)method. Results:Traditional Chinese medicine high dose group,western medicine group,FS and EF values and model groups increased significantly(P〈0.01),traditional Chinese medicine high dose group,western medicine group rats FS,EF value and normal group had no statistical significance(P〉0.05);model group ST2,IL-33 levels were significantly increased(P〈0.01),traditional Chinese medicine high dose group,middle dose group and Western medicine group ST2,IL-33 levels decreased significantly(P〈0.01),western medicine group and traditional Chinese medicine high dose group,middle dose group,the difference was not statistically significant(P〉0.05),traditional Chinese medicine high dose group,middle dose group of IL-33 levels compared with the normal group had no statistical significance(P〉0.05). Conclusion:Zhenwu Decoction can significantly reduced heart failure rat serum ST2,IL-33 levels,that Zhenwu Decoction by regulation IL-33/ST2 L signaling pathway improve ventricular reconstruction,which may is one of the mechanisms of Zhenwu Decoction in the treatment of heart failure.
