摘要 目的研究姜黄素对人肺腺癌NCI—H1975细胞放射增敏的作用。方法选取人肺腺癌NCI-H1975细胞作为研究对象,将细胞分为对照组、单独姜黄素处理组、单独放射处理组、姜黄素联合放射处理组。采用CCK-8实验检测细胞增殖的变化;流式细胞仪检测细胞周期的改变;细胞核Hochest33258染色技术和AnnexinV—FITC/PI双染技术检测细胞凋亡的情况。结果CCK-8实验结果发现姜黄素与X射线联合处理能显著抑制肺腺癌细胞增殖,且具有浓度依赖性。流式细胞仪检测发现姜黄素与X射线联合作用可以将细胞大部分阻滞在S期。细胞核染色及AnnexinV—FITC/PI双染技术结果发现姜黄素联合x射线可以增强姜黄素诱导的细胞凋亡作用。结论姜黄素联合X射线对人肺腺癌NCI-H1975细胞有很好的增殖抑制效果,其机制可能与细胞的S期阻滞有关,并且可以诱导细胞凋亡。 Objective To study the sensitization effect of curcumin on lung cancer NCI-H1975 cell. Methods The NCI-H1975 cells were randomly divided into four groups:control group, curcumin group, X-ray group, curcurnin+ X-ray group. Cell growth suppression of each group was analyzed by CCK-8 assay. The cell cycle and apoptosis of each group were detected by flow cytometry and Hochest33258 staining. Results The proliferation of NCI-H1975 cells was inhibited remarkably in curcumin+ X-ray group,which was concentration-dependent. Curcumin combined X-ray could result in S phase arrest of cells, and induce cell apoptosis. Conclusions Curcumin combined X-ray can radiosensitize the NCI-H1975 cell, which can induce apoptosis, and S phase arrest may be one of the mechanisms.
