摘要
目的探讨邻苯二甲酸单乙基己基酯(MEHP)对中国仓鼠卵巢细胞(CHO细胞)的细胞毒性和对凋亡基因和癌基因表达水平的影响。方法采用4个不同剂量的MEHP对CHO细胞染毒24 h后,采用荧光定量PCR检测4个凋亡基因Bcl-2、caspase-3、caspase-9,Bax及癌基因p53表达水平的改变。结果 0.5 mmol/L MEHP染毒时caspase-3、caspase-9、Bax表达水平较对照组显著增加,差异均有统计学意义(P<0.01或P<0.05);凋亡基因Bcl-2表达水平在4个剂量组均比对照组显著降低(P<0.05或P<0.01),在0.25与0.5 mmol/L MEHP作用下,Bax与Bcl-2比值显著降低(P<0.05或P<0.01),p53在0.25和0.5 mmol/L MEHP浓度下表达下降。结论 MEHP可通过激活线粒体caspase凋亡信号通路引起CHO细胞凋亡,同时可引起抑癌基因表达水平下降。
[Objective]To observe the toxic effects of mono-2-ethylhexyl phthalate(MEHP) on Chinese hamster ovary(CHO)cells and detect the influence of on expression level of apoptosis genes and oncogenes.[Methods]CHO cells were treated with different doses of MEHP for 24 hours,the expression levels of four apoptosis genes(Bcl-2,caspase-3,caspase-9,Bax) and oncogene p53 were determined by real-time fluorescence quantitative PCR.[Results]In 0.5 mmol/LMEHP exposure group,the expression levels of caspase-3,caspase-9 and Bax were higher than those in the control group,and the differences were statistically significant(P〈.01 or P〈.05).The expression levels of Bcl-2 in four exposure groups were significantly lower than that in the control group(P 0.01 or P 0.05).The ratio of Bax to Bcl-2 decreased in 0.25 mmol/L and 0.5 mmol/L MEHP exposure groups(P〈.05 or P〈.01),and the expression level of p53 in 0.25 mmol/L and 0.5 mmol/L MEHP exposure groups decreased.[Conclusion]MEHP could induce apoptosis in CHO cells through mitochondria caspase pathway activation,and antioncogene expression significantly decreases after DEHP treatment in CHO cells.
