摘要
目的探讨新疆汉族T2DM及糖耐量正常(NGT)人群miR-375启动子区部分CpG位点的甲基化状态。方法采用基质辅助激光解析电离时间飞行质谱仪检测100例T2DM患者(T2DM组)和100名NGT者(NGT组)外周血白细胞中miR-375基因启动子区5个CpG位点的甲基化状态,并对结果进行分析。结果 (1)T2DM组平均甲基化水平高于NGT组(10.27%vs 7.24%,P<0.01);(2)T2DM组miR-375基因5个CpG位点的甲基化率均高于NGT组,如CpG_22、CpG_24.25、CpG_26.27,仅CpG_26.27在两组中比较,差异有统计学意义(P<0.01)。结论汉族T2DM患者外周血白细胞中miR-375基因部分启动子区异常甲基化水平可能与T2DM的发生有一定的关系。
Objective to compare the methylation status ot UpG sites in miR-375 promoter region in different glucose tolerance Han people in Xinjiang area.Methods 100 patients with type 2 diabetes(T2DM group) and 100 healthy volunteers(NGT group) were recruited in this study.The methylation levels of CpG five sites of miR-375 promoter in leukocytes were determined by MassARRAY spectrometry.Results(1) The mean levels of miR-375 methylation were higher in T2 DM group than that in NGT group(10.27%vs 7.24%,P〈0.01).(2)The level of MiR-375 gene methylation in each CpG site(CpG22,CpG23.24 and CpG25.26) was higher in T2 DM group than that in NGT group,with significant difference for CpG25.26(P〈0.01).Conclusion The methylation of CpG sites of miR-375 gene promoter in peripheral blood leukocytes may be related with the occurrence of T2 DM.
