摘要
建立了以C18反相柱为分析柱,以甲醇-水为流动相的反相液相色谱法,采用1 mL/min的流速在检测波长为280 nm处对小米中α-生育酚的含量进行测定。结果表明:α-生育酚在质量浓度为0.5—100μg/mL范围内线性良好,R^2=0.9993,回收率83%—99%。在选定的5种小米中,α-生育酚含量各异,可能与品种有关。该方法操作简便,结果准确,可用于小米α-生育酚的含量测定。
A high performance liquid chromatographic (HPLC)method with C18 reversed phase column as the analytical column and methanol-water as the mobile phase was established. Theα-tocopherol content in millet was determined at 280 nm wavelength with flow rate of 1 mL/min. The results showed that α-tocopherol had a good linearity in the range of 0.5--100μg/mL with R2 =0. 9993 ,and the recovery rate was 83%--99%. There were different α-tocopherol contents in the selected 5 varieties of millet, which may be related to the variety. This method is simple, accurate, and can be used for determination of α-tocopherol content in millet.
