摘要
目的探讨正常大鼠苍白球orexin-1(OX1)受体的表达,以及苍白球微量注射OX1受体激动剂orexin-A和阻断剂SB-334867对大鼠运动行为的影响。方法采用免疫组织化学方法检测正常大鼠苍白球OX1受体的表达,采用核团埋管术和提升躯体摇摆实验观察orexin-A和SB-334867对大鼠运动行为的影响。结果免疫组织化学染色显示,大鼠苍白球神经元表达OX1受体。单侧苍白球微量注射orexin-A可使大鼠发生明显对侧摇摆,单侧苍白球微量注射SB-334867可使大鼠发生明显的同侧摇摆,与注射生理盐水大鼠比较,差异均有统计学意义(F=132.7,t=8.57、9.43,P<0.01)。结论大鼠苍白球表达OX1受体,苍白球orexin-A可能通过激活OX1受体调节机体运动功能。
Objective To observe the expression of orexin-1 (OX1) receptors in globus pallidus of normal rats, and the effects of microinjection of OX1 receptor agonist orexin-A and blocking agent SPr334867 on rat locomotor activity. Methods Employing immunohistochemical staining, the expression of OX1 receptors in globus pallidus of normal rats was detected, and applying nuclear group buried tube and lifting body sway test, the impacts of orexin-A and SB-334867 on locomotor activity of rat were observed. Results Immunohistochemical staining indicated that OX1 receptors expressed in neurons of the globus pallidus. Unilateral microinjection of orexin-A into the globus pallidus might cause contralateral sway in the rat, and unilateral microinjeetion of SB-334867 into globus pallidus could make the rat ipsilateral swing, compared with those injected with normal saline, the diffe fences being significant (F=132.7;t=8.57,9.43,P〈0.01). Conclusion OX1 receptors express in the globus pallidus of rats. Pallidal orexin-A regulates the body movement function is possibly through activating OX1 receptors.
