摘要
目的探讨MicroRNA-21在胃癌细胞上皮-间质转化中的作用及其机制。方法利用Lipofectamine 2000脂质体将miR-21 mimics、miR-21 inhibitor转染入AGS细胞中;Real-time聚合酶链反应(RT-PCR)法测定AGS细胞内miR-21的表达;采用划痕法、Transwell小室检测细胞的迁移和侵袭能力;Western blotting方法检测上皮-间质转化(EMT)相关蛋白表达的表达变化。结果 AGS细胞转染miR-21 mimics后,细胞的迁移和侵袭能力均高于对照组(P<0.05),Western blot结果显示E-Cadherin、PTEN表达下降,Vimentin表达升高;AGS细胞转染miR-21 inhibitor后,细胞的迁移和侵袭能力均低于对照组(P<0.05);E-Cadherin、PTEN表达升高,Vimentin表达降低。结论 miR-21可以诱导AGS胃癌细胞迁移和侵袭。E-cadherin和PTEN表达下调和上调Vimentin可能是miR-21作用的分子机制。miR-21在胃癌肿瘤转移中可能起着重要作用,可能成为胃癌治疗的一个潜在靶点。
Objective in gastric carcinoma cells. To investigate the role of miR-21 in the process of epithelial-mesenchymal transition (EMT) Methods AGS cells were transfected with miR-21 mimics and miR-21 inhibitor. Real-time PCR was used to measure miR-21 mRNA expression. E-Cadherin, Vimentin and PTEN proteins was detected using western blotting. The ability in invasiveness and migration was evaluated using transwell and wound-healing assay, respectively. Results Up-regulation of miR-21 induced increasing protein expression of Vimentin and decreasing expression of E-Cad- herin and PTEN in AGS gastric cancer cells as well as the increase in cellular invasiveness and migration. Down-regulation of miR-21 showed the opposite biological effects. Conclusion MiR-21 induced invasiveness and migration in AGS gastric cancer cells. The down-regulation of E-Cadherin and PTEN and up-regulation of Vimentin might be the underlying molecular mechanism. MiR-21 might play an important role in tumor metastasis in gastric cancer as a potential target for treatment of gastric cancer.
