针刺对糖尿病大鼠胰腺内质网应激PERK-CHOP途径与Bax/Bcl-2基因表达的影响 被引量:15

Effects of acupuncture on pancreatic ERS PERK-CHOP pathway and Bax/Bcl-2 mRNA expressions in diabetic rats

摘要 目的:探讨内质网应激途径PERK-CHOP与链脲佐菌素(STZ)诱导的糖尿病大鼠胰腺相关凋亡基因表达的关系及针刺对其的影响。方法:70只雄性SD大鼠高脂高糖饲料喂养4周后腹腔注射小剂量STZ(30mg·kg^(-1)·d^(-1)),成功制造糖尿病模型30只,随机分为模型组、针刺组和二甲双胍组,每组10只。治疗4周后,实时荧光定量PCR测胰腺CHOP、PERK、Bax、Bcl-2 mRNA表达,并设空白组对照。结果:与治疗前比较,治疗后针刺组和二甲双胍组RBG均明显降低(P<0.05);干预4周后,针刺组和二甲双胍组RBG仍显著高于空白组(P<0.01),虽低于模型组,但差异无统计学意义。干预4周后,模型组胰腺CHOP、PERK和Bax mRNA表达均显著高于空白组(P<0.01),针刺组和二甲双胍组均显著低于模型组(P<0.01);模型组Bcl-2 mRNA表达显著低于空白组(P<0.01),针刺组和二甲双胍组显著高于模型组(P<0.01)。结论:STZ诱导的糖尿病大鼠胰腺组织存在内质网应激和细胞凋亡,针刺可以通过PERK-CHOP通路抑制胰腺内质网应激,改善凋亡基因表达。 Objective: To investigate the relationship between pancreatic endoplasmic reticulum stress(ERS) PERKCHOP pathway and cell apoptosis-related gene expression in diabetic rats(DM) and the influence of acupuncture on it. Methods: A total of 70 male SD rats were fed with high fat and high glucose food for 4 weeks and were intraperitoneal injected with lowdose STZ(30mg·kg^(-1)·d^(-1)). Thirty DM model rats were divided randomly into model group(MG), acupuncture group(AG) and metformin group(MFG) with 10 rats in each group. The mRNA expression of CHOP, PERK, Bax and Bcl-2 was measured by real-time fluorescent PCR after 4 weeks of treatment compared with the control group(CP). Results: Compared with the outcomes before treatment, the RBG of AG and MFG were significantly decreased(P<0.05). After 4 weeks of treatment, the RBG in AG and MFG were significantly higher than CG(P<0.01), and lower than MG, but the difference was not statistically significant. the pancreatic mRNA expressions of CHOP, PERK and Bax in MG were significantly higher than CG(P<0.01), the mRNA expressions of CHOP, PERK and Bax in AG and MFG were significantly lower than MG(P<0.01) and higher than CG(P<0.01). The mRNA expression of Bcl-2 in MG was lower than CG(P<0.01). The mRNA expression of Bcl-2 in AG and MFG was significantly higher than MG(P<0.01). Conclusion: STZ-induced DM rats manifest pancreatic ERS and cell apoptosis, and acupuncture could repress pancreatic ERS and improve cell apoptosis- related gene expressions by PERK-CHOP pathway.
