铁皮石斛醇提不同萃取物对转化生长因子β1诱导的HSC-T6细胞的影响 被引量:4

Effects of different extractant of methanol extract of Dendrobium officinale on TGFβ1-induced HSC-T6 cell line

摘要 目的:探讨铁皮石斛醇提不同萃取物对转化生长因子β1(TGFβ1)诱导的HSC-T6细胞增殖及相关细胞因子的影响。方法:建立TGFβ1诱导HSC-T6细胞增殖模型,MTT法探讨萃取物对细胞增殖的影响;并采用RTPCR、ELISA法测定其对细胞相关因子的影响。结果:MTT结果表明,正己烷层、氯仿层、乙酸乙酯层对TGFβ1诱导的HSC-T6细胞增殖有显著的抑制作用(P<0.05)。RT-PCR、ELISA结果显示,与模型组比较,正己烷层显著降低α-SMA、CTGF、TβR-Ⅱ的m RNA含量和蛋白表达(P<0.05),氯仿层能抑制CTGF、TβR-Ⅱ的m RNA含量和蛋白表达(P<0.05),乙酸乙酯层组抑制CTGF的m RNA和蛋白表达(P<0.05)。结论:铁皮石斛醇提萃取物中的正己烷层可能通过抑制α-SMA、CTGF、TβR-Ⅱ抑制肝星状细胞的活化,氯仿层、乙酸乙酯层可能通过抑制CTGF、TβR-Ⅱ抑制肝星状细胞的活化。 Objective: To investigate the effects of the different extractant of methanol extract of Dendrobium officinale on the proliferation and expression of cytokines of TGFβ1-induced HSC-T6 cell line. Methods: MTT method was used to evaluate the effects of different extractant of methanol extract of Dendrobium officinale on TGFβ1-induced HSC-T6 cells proliferation. A fluorescence RT-PCR and ELISA assay were used to assess the effects of each layer of methanol extracts on expression of associated cytokines in HSC-T6 cell. Results: MTT assay results showed that n-hexane, chloroform, ethyl acetate had an inhibitory effects on the proliferation of TGFβ1-induced HSC-T6 cells(P<0.05). RT-PCR, ELISA assay results showed that compared with the model group, n-hexane group reduced the content ofα-SMA, CTGF, TβR-Ⅱ(P<0.05), chloroform, ethyl acetate reduced the content of CTGF, TβR-Ⅱ(P<0.05). Conclusion: N-hexane extract is able to inhibit the proliferation of HSC-T6 may be through inhibiting the expression of α-SMA, CTGF, TβR-Ⅱ, while chloroform extract and ethyl acetate extract can inhibit the proliferation of HSC-T6 may be via inhibiting the expression of CTGF, TβR-Ⅱ.
