基于网络药理学研究龙生蛭胶囊抗脑卒中的作用机制 被引量:14

Mechanism of Longshengzhi Capsule in the treatment of stroke based on network pharmacology

摘要 目的:采用网络药理学的方法,研究龙生蛭胶囊活性成分、作用靶点及生物学意义,探讨其抗脑卒中的作用机制。方法:采用TCMSP数据库、台湾中医药数据库、BATMAN-TCM等数据库按照龙生蛭胶囊不同药物组分获取龙生蛭胶囊的成分,通过Swiss Target Prediction数据库、TTD数据库及Drugbank数据库进行靶点的预测和收集;通过OMIM、TTD、Genecards、DiGSeE、CTD等5个疾病相关靶点的数据库检索脑卒中相关靶点信息。将药物预测靶点映射至疾病靶点集合上,获得交集靶点,运用DAVID数据库对得到的交集进行GO和pathway的富集分析,通过cytoscape软件对结果进行可视化呈现。结果:网络分析结果发现,龙生蛭胶囊主要通过干预脑卒中过程中炎症过程、营养因子分泌及免疫相关淋巴细胞的调节等过程,来缓解脑卒中所造成的损伤。其中,AKT1、JUN、MAPK14、MAPK8、RELA等为关键的调节位点。结论:基于网络药理学的分析,龙生蛭胶囊能从调节炎症过程、营养因子分泌及免疫相关淋巴细胞调节等方面,对脑卒中的病理进行调节。 Objective:To study the active components,target and biological significance of Longshengzhi Capsule by network pharmacology,and to explore its anti-stroke mechanism.Methods:The TCMSP database,Taiwan Residents Chinese medicine database,BATMAN-TCM database were used to obtain the components of Longshengzhi Capsule according to the different therapeutic groups.The targets were predicted and collected through the Swiss Target Prediction database,TTD database and Drugbank database.The target information of stroke-related targets was searched through a database of five disease-related targets database,including OMIM,TTD,Genecards,DiGSeE,and CTD.The drug predicted targets was mapped to the disease target set to obtain the intersection target.The DAVID database was used to enriched the intersection genes via GO and pathway methods,and the results were visualized by cytoscape software.Results:The results of network analysis found that Longshengzhi Capsule majorly relied on the process of inflammatory process,trophic factor secretion and immune-related lymphocyte regulation during stroke to alleviate the damage caused by stroke.Among the selected genes,AKT1,JUN,MAPK14,MAPK8 as well as RELA were regarded as the hub genes.Conclusion:Based on the analysis of network pharmacology,Longshengzhi Capsule can regulate the pathological process of stroke from the aspects of regulating inflammation process,trophic factor secretion and immune-related lymphocyte regulation.This present study also provides a theoretical basis to further illustrate the mechanism of Longshengzhi Capsule in the improvement of the situation of stroke patients.
