菟丝子提取物通过BMP2/Smad4信号通路对MC3T3-E1细胞增殖的影响 被引量:19

Effects of the extract of dodder seed on the proliferation of MC3T3-E1 cells through the BMP2/Smad4 signaling pathway

摘要 目的:研究中药菟丝子对MC3T3-E1细胞促增殖作用及其可能的作用机制。方法:体外培养小鼠成骨细胞MC3T3-E1,通过MTT法检测菟丝子提取物对细胞增殖的影响,通过相关试剂盒检测碱性磷酸酶(ALP)活性和钙盐沉积水平,Western Blot检测Ⅰ型胶原蛋白、BMP2和Smad4蛋白表达。结果:菟丝子提取物能够促进小鼠成骨细胞MC3T3-E1增殖,提高ALP活性和钙盐沉积,并上调Col-Ⅰ、BMP2和Smad4蛋白表达(P<0.05),菟丝子提取物浓度在10μg/mL时促进作用较明显。结论:菟丝子提取物能够促进成骨细胞增殖,其作用机制可能与调节BMP2/Smad4信号通路有关。 Objective:To study the proliferation-promoting effects of the extract of dodder seed on MC3 T3-E1 cells and its possible mechanism.Methods:Mouse osteoblasts MC3 T3-E1 cells were cultured in vitro,and the effects of the extract of dodder seed on MC3 T3-E1 cell proliferation was detected by MTT method.Alkaline phosphatase(ALP)activity and the level of calcium salt deposition were detected by relevant kits.The protein expression of typeⅠcollagen,BMP2 and Smad4 was analyzed by western blot.Results:The extract of dodder seed could promote the proliferation of MC3 T3-E1 cells,enhanced the activity of ALP and calcium salt deposition,and up-regulated expression of Col-Ⅰ,BMP2 and Smad4 protein,the difference was statistically significant(P<0.05).Conclusion:The extract of dodder seed promotes proliferation of osteoblast cells and its mechanism may be related to the regulating BMP2/Smad4 signal pathway.
